藏法合成人工牛黄促进糖尿病创伤愈合的机制研究

摘要 目的评价藏法合成的人工牛黄促糖尿病创伤愈合的作用,并探讨其机制。方法通过RAW264.7细胞炎症模型,观察藏法人工牛黄对抗炎因子表达的影响。建立糖尿病大鼠创伤模型,评价藏法人工牛黄对糖尿病创伤的治疗效果。结果藏法人工牛黄在体外可显著促进RAW264.7炎症细胞中白细胞介素(IL)-10mRNA的表达,体内可显著促进创伤组织中IL-10和精氨酸的表达,增加创伤处M2型巨噬细胞的数量,降低IL-1β、肿瘤坏死因子α的表达,促进糖尿病创伤愈合。结论藏法人工牛黄可能是通过促进糖尿病创伤部位巨噬细胞向M2型转化、分泌抗炎因子并抑制促炎症因子的表达来促进创伤的愈合。研究结果为藏法人工牛黄在慢性创伤临床治疗上的应用提供了依据。 OBJECTIVE To evaluate the effect of artificial bezoar synthesized by Tibetan method on diabetic wound healing,and to explore its mechanism.METHODS RAW264.7 cells were used to evaluate the effect of Tibetan artificial bezoar on the expression of anti-inflammatory factors.The therapeutic effect of Tibetan artificial bezoar on diabetic trauma was evaluated by establishing a diabetic rat trauma model.RESULTS Tibetan artificial bezoar significantly promoted the expression of IL-10 mRNA in RAW264.7 inflammatory cells.In vivo,Tibetan artificial bezoar significantly promoted the expression of IL-10 and arginine in the wound tissue,promoted the infiltration of M2 macrophages in the wound tissue,and reduced the expression of IL-1βand TNF-α,thereby significantly promoting the healing of diabetic wound.CONCLUSION The Tibetan artificial bezoar may promote the healing of diabetic wound by promoting the transformation of macrophages into M2 type,secreting anti-inflammatory factors such as IL-10 and arginine,and inhibiting the expression of pro-inflammatory factors IL-1β and TNF-α.This study provides a theoretical basis for the application of Tibetan artificial bezoar in the clinical treatment of chronic trauma.
