HPLC测定姜黄鲜切饮片中的姜黄素类成分

Determination of curcumin class content of freshly cut decoction pieces of Curcumae LongaeRhizoma

摘要 目的以乙醇-水为洗脱系统来测定姜黄鲜切饮片中姜黄素类成分的含量。方法采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙醇-水(41:59),流速0.75 mL·min^(-1),柱温35℃,检测波长430 nm。结果姜黄素、去甲氧基姜黄素、双去甲氧基姜黄素的线性范围为32.4~486.0、6.2~93.0、17.8~267.0μg·mL^(-1)(r^(2)均≥0.9998);平均回收率为100.97%~103.39%,RSD为0.96%~1.69%;供试品溶液在24 h内稳定,RSD为0.15%~0.83%(n=7)。11批样品中姜黄素、去甲氧基姜黄、双去甲氧基姜黄素的含量分别为1.76%~2.49%、0.53%~0.76%、0.43%~0.63%。结论所用方法结果可靠、环保、准确、重复性好、回收率高,可用于姜黄鲜切饮片的含量测定。 OBJECTIVE To determine the content of curcumin class in freshly cut decoction pieces of Curcumae Longae Rhizoma by HPLC with ethanol-water.METHODS The determination was performed on a Kromasil C_(18)(250 mm×4.6 mm,5μm)column with a mobile phase gradient elution of ethanol-water(41:59).The flow rate was 0.75 mL·min~(-1),column temperature was 35℃,and the detection wavelength was 430 nm.RESULTS A good linear relationship was obtained between the peak areas and the concentrations of curcumin,desmethoxycurcumin and bisdesmethoxycurcumin in the range of 32.4-486.0μg·mL^(-1),6.2-93.0μg·mL^(-1),and 17.8-267.0μg·mL^(-1).The average recovery was in the range of 100.97%-103.39%,with the RSD of 0.96%-1.69%.The test solution was stable within 24 h with the RSD of 0.15%-0.83%(n=7).The content of curcumin,desmethoxyturmeric and bisdesethoxycurcumin in 11 batches of samples were 1.76%-2.49%,0.53%-0.76%,and 0.43%-0.63%,respectively.CONCLUSION The method is reliable,environmentally friendly,accurate,and repeatable with high recovery,which can be used for determination of curcumin class content in freshly cut decoction pieces of Curcumae Longae Rhizoma.
