基于小鼠模型评估市售宣称敏感性皮肤适用润肤剂的温和性

Evaluation of the Mildness of Commercially Available Moisturisers Claiming to be Suitable for Sensitive Skin Based on a Murine Model

摘要 利用小鼠皮肤较薄的特点,研究市售敏感性皮肤适用润肤剂的温和性。收集10款宣称敏感性皮肤适用的润肤剂,先利用家兔多次皮肤刺激性试验以及重组人表皮模型评价受试产品潜在的皮肤刺激性。再进行C57小鼠涂抹试验,每天2次连续7天,然后检测经皮水分丢失(TEWL)、皮肤表面pH值以及角质层含水量等屏障功能参数。结果表明所有受试产品在家兔和重组人表皮模型上均未显示明显的皮肤刺激性,但是其中5款产品可在小鼠模型中诱导表皮功能异常,显著降低了小鼠皮肤角质层含水量或明显增加TEWL。建议在敏感性皮肤适用产品的研发阶段增加小鼠经皮暴露试验帮助筛选温和的配方。 The mildness of commercial available moisturisers intended for individuals with sensitive skin was evaluated by studying the potential impacts on the thin skin of mice.Ten commercial moisturisers claiming to be“suitable for people with sensitive skin”were collected,and their skin irritation potential was evaluated using the rabbit skin irritation assay and a reconstructed human epidermis model.Futhermore,following topical application of the products on the back of C57 mice twice daily for 7 days,and then skin barrier function parameters such as transepidermal water loss(TEWL),skin surface pH and stratum corneum(SC)hydration were tested.It was shown that none of the products caused significant skin irritation in rabbits or changed the tissue cell viability of the recombinant human epidermal models, but 5 of them could induce abnormalities in epidermal function in murine models, coupled with elevations in TEWL and reduced SC hydration. It is suggested that the murine model could be involved to predict the mildness and applicability of skin care products designed for people with sensitive skin before launching.
