摘要
目的:利用生物信息学方法分析角鲨烯环氧化酶(SQLE)在软组织肉瘤(SARC)中的表达、预后及其作用机制。方法:下载UCSC数据库中泛癌的表达及预后数据,分析SQLE在泛癌中的表达及预后。分析SQLE在GTEx正常组织表达与SARC肿瘤组织表达的差异,然后通过单因素和多因素分析SQLE在SARC中的预后意义。探讨SQLE表达与SARC中免疫浸润细胞的关系,并通过基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析探讨SQLE在SARC中的潜在机制。结果:SQLE在多种肿瘤中异常表达,并且与多种肿瘤的总体生存率(OS)和无进展生存期(PFS)明显相关。与GTEx中正常软组织比较,SQLE在SARC中表达明显升高(P<0.001),并且在每种亚型中均表达升高(P<0.001)。生存分析结果显示,SQLE高表达的SARC患者有更差的OS(P<0.001)和PFS(P<0.01)。单因素分析和多因素分析结果显示,SQLE在SARC中是OS(P<0.001)和PFS(P<0.05)的独立风险因素。肿瘤浸润免疫细胞分析发现,SQLE表达与SARC中休眠树突细胞、嗜酸性粒细胞、M0巨噬细胞、休眠肥大细胞、单核细胞、CD8T淋巴细胞6种免疫细胞明显相关。结论:SQLE在多种肿瘤中表达升高,包括SARC。SQLE表达与SARC的差预后相关,是不良预后的独立风险因素。
Objective To explore the expression,prognostic role and potential mechanism of squalene epoxidase(SQLE)in soft tissue sarcoma by bioinformatic analysis.Methods The expression and prognostic role of SQLE in pan-cancer were analyzed via the expression and prognostic data from UCSC database.The expression of SQLE in soft tissue sarcoma(SARC)tissues and GTEx normal tissues was analyzed.Univariate and multivariate Cox analysis were performed to explore the correlation between SQLE expression and survival in SARC.Then we explored the correlation between SQLE expression and Tumor-infiltrating immune cells in SARC.In addition,the potential mechanism of SQLE was analyzed by gene ontology(GO)enrichment and kyoto encyclopedia of genes and genomes(KEGG)pathway analysis.Results Pan-cancer analysis showed that SQLE expression was abnormally and associated with overall survival(OS)and progression-free survival(PFS)in a variety of tumors.compared to normal soft tissues in GTEx,SQLE expression was significantly elevated in SARC(P<0.001)and was elevated in every subtype(P<0.001).Survival analysis showed that SARC patients with high SQLE expression had worse OS(P<0.001)and PFS(P<0.01).Univariate and multivariate Cox analyses showed that SQLE was an independent risk factor for OS(P<0.001)and PFS(P<0.05)in SARC.Analysis of tumor-infiltrating immune cells showed that SQLE expression was significantly correlated with DC resting,eosinophils,M0,mast cells resting,monocytes,and T cells CD8 immune cells in SARC.Conclusion SQLE expression was elevated in a variety of tumors,including SARC.SQLE expression was associated with poor prognosis of SARC,was an independent risk factor for poor prognosis,and can be used as a biomarker of prognosis.
