摘要
细胞焦亡是由Gasdermines(GSDMs)介导的一种程序性细胞死亡.GSDMs的N端结构域在质膜上形成孔隙,使细胞膜破裂和胞内容物释放导致炎性反应,介导细胞焦亡.细胞焦亡在炎症性疾病及恶性肿瘤中都起着重要作用.随着细胞焦亡的深入研究,越来越多的研究结果显示细胞焦亡途径能够调控结直肠癌的肿瘤微环境和抗肿瘤免疫,与结直肠癌的发生发展、治疗预后有着密切的关系.本文通过介绍焦亡的分子机制,探讨细胞焦亡在结直肠癌发生发展、免疫微环境以及治疗方面的作用,以期为结直肠癌临床诊疗提供新的见解,为开发基于细胞焦亡途径的抗结直肠癌治疗靶点提供理论依据.
Pyroptosis is a type of programmed cell death mediated by Gasdermines(GSDMs).The N-terminal structural domains of GSDMs form pores in the plasma membrane,causing rupture of the cell membrane and release of intracellular contents leading to an inflammatory response that mediates pyroptosis.Pyroptosis plays an important role in both inflammatory diseases and malignant tumors.With the in-depth study of pyroptosis,more and more studies have found that the pyroptosis pathway can regulate the tumor microenvironment and anti-tumor immunity in colorectal cancer,which is closely related to the development and prognosis of colorectal cancer treatment.In this review,we introduce the molecular mechanism of pyroptosis and discuss the role of pyroptosis in colorectal cancer development,immune microenvironment and treatment,with the aim of providing new insights into the clinical diagnosis and treatment of colorectal cancer and the theoretical basis for the development of anti-tumor therapeutic targets based on the pyroptosis pathway.
