5-氨基乙酰丙酸采用醇质体包载前后的稳定性

摘要 研究5-氨基乙酰丙酸(5-ALA)被醇质体包载前后在不同介质pH、温度和光线条件下的稳定性。以大豆卵磷脂和胆固醇为囊材,采用乙醇注入法制备5-ALA醇质体;以5-ALA相对含量为指标,对比分析原料药和醇质体中药物的稳定性变化规律。结果表明,5-ALA醇质体中药物的包封率为85.21%,介质pH、温度和光线等因素均会影响5-ALA的稳定性,在碱性条件下5-ALA更容易降解,温度越高降解速度越快,光照对药物的降解影响相对较小,5-ALA被醇质体包载前后可有效提高其稳定性。 The stability of drugs before and after being encapsulated by ethosomes was studied under different pH media,temperature,and light conditions.Using soybean lecithin and cholesterol as capsule materials,5-aminolevulinic acid hydrochloride(5-ALA)liposomes were prepared by ethanol injection method.Using the relative content of 5-ALA as an indicator,the stability changes of drugs in bulk drugs and ethosomes was compared and analyzed.The results showed that the encapsulation efficiency of the drug in 5-ALA liposomes was 85.21%.Factors such as medium pH,temperature,and light could all affect the stability of 5-ALA.Under alkaline conditions,5-ALA was more easily degraded,and the degradation rate was faster with higher temperature.The effect of light on drug degradation was relatively small,and the stability of 5-ALA could be effectively improved after being encapsulated by ethosomes.
