松乳菇多糖提取工艺研究及降血糖活性评价 被引量:1

摘要 以松乳菇为研究对象,选择不同提取方法对松乳菇多糖进行提取,通过单因素试验确定影响多糖提取率的主次因素,并通过正交试验优化提取工艺。最终确定纤维素酶酶提法提取松乳菇多糖效果好,影响多糖提取率的主次因素为提取温度>酶用量>提取时间,最佳提取工艺为酶用量30 mg、提取时间2.0 h、提取温度70℃。试验采用α-淀粉酶和α-葡萄糖苷酶的活性为指标,测定各多糖样品的体外降血糖活性。在最佳提取条件下得到的松乳菇提取物中多糖含量最高,含量达到45.70%,且对α-淀粉酶活性抑制和α-葡萄糖苷酶活性抑制作用较强,抑制率可达41.80%和41.56%。 Lactarius deliciosus was used as the research object,and different extraction methods were selected to extract polysaccharides from L.deliciosus.The primary and secondary factors affecting the polysaccharides extraction rate were determined by single factor experiment,and the extraction process was optimized by orthogonal experiment.Finally,it was determined that cellulase was the best method to extract L.deliciosus polysaccharides.The main and secondary factors affecting the extraction rate were extraction temperature>enzyme dosage>extraction time,and the best extraction process was 30 mg enzyme dosage,2.0 h extraction time and 70°C extraction temperature.The hypoglycemic activity of each polysaccharide sample in vitro was determined by the activity ofα-amylase andα-glucosidase.The polysaccharide content of the L.deliciosus extract obtained under the optimal extraction conditions was the highest,reaching 45.70%,and the inhibition ofα-amylase activity andα-glucosidase activity was strong,with inhibition rates of 41.80%and 41.56%.
