幽门螺杆菌小RNA调控毒力基因的作用与机制

摘要 幽门螺杆菌(Hp)是导致胃部疾病的主要危险因素之一,可通过调节毒力基因的表达适应环境并感染宿主。由于Hp基因组编码的转录调节因子较少,转录后调控在调节Hp基因表达中发挥重要作用。小RNA(sRNA)由非编码RNA基因产生,是一类长度为50~400个核苷酸的RNA分子,可通过与靶信使RNA(mRNA)结合抑制翻译或影响核糖核酸酶对mRNA的降解,从而调控基因表达。Hp含有数百个sRNA,参与调控毒力基因表达,影响细菌的致病性。因此,深入研究Hp sRNA对毒力基因的调控作用与机制,有助于阐明Hp sRNA在Hp菌株毒力差异中的作用及菌株间致病性的差异。 Helicobacter pylori(Hp)is one of the most important risk factors for gastric diseases,which could regulate the expression of related virulence genes to fit the host environment and infect the host.Few transcriptional regulators are encoded by Hp genome and post-transcriptional regulation has a major control of Hp gene expression.Small RNA(sRNA)is a category of 50-400 nt long RNA molecules produced by non-coding RNA genes.They bind to target messenger RNA(mRNA)inhibiting translation or impacting the degradation of mRNA by RNase,thus regulating the gene expression.Hundreds of sRNA have been found in Hp,involved in regulating expression of virulence genes and influencing pathogenicity.Therefore,by studying the role and mechanism of Hp sRNA in regulating virulence gene,it is excepted to illuminate the roles of Hp sRNA in virulent difference of Hp strains and differences in pathogenicity between strains.
