SULT2A1、VEGF-C、IGF-Ⅰ在子宫肌瘤患者中的表达及相关性研究

摘要 目的探讨硫酸基转移酶2A1(SULT2A1)、血管内皮生长因子-C(VEGF-C)、胰岛素样生长因子-Ⅰ(IGF-Ⅰ)在子宫肌瘤患者中的表达及相关性。方法选取2020年12月至2022年12月江苏大学附属宜兴市人民医院病理科保存的50例女性子宫肌瘤组织(子宫肌瘤组织组)及瘤旁组织(瘤旁组织组)标本,并选取同时期同一医院病理科保存的65例正常子宫肌组织(正常子宫肌组织组)作为研究对象。采用免疫组化法检测各标本SULT2A1、VEGF-C蛋白,采用流式细胞术检测IGF-Ⅰ的表达;绘制受试者工作特征(ROC)曲线分析SULT2A1、VEGF-C、IGF-Ⅰ单独检测及联合检测对子宫肌瘤的诊断价值;分析SULT2A1、VEGF-C、IGF-Ⅰ表达之间的相关性。结果相比于正常子宫肌组织及瘤旁组织,子宫肌瘤组织VEGF-C、IGF-Ⅰ表达较高,SULT2A1表达较低(P<0.05)。瘤旁组织VEGF-C、IGF-Ⅰ表达高于正常子宫肌组织,SULT2A1低于正常子宫肌组织(P<0.05)。相关性分析显示,SULT2A1和VEGF-C表达呈负相关(r=-0.902,P=0.001),IGF-Ⅰ和VEGF-C表达呈正相关(r=0.860,P=0.001),SULT2A1和IGF-Ⅰ表达呈负相关(r=-0.854,P=0.001);绘制ROC曲线显示,VEGF-C、IGF-Ⅰ、SULT2A1单独预测价值低于三项联合。结论VEGF-C、IGF-Ⅰ在子宫肌瘤组织中高表达,SULT2A1在子宫肌瘤组织中低表达,三者具有相关性,VEGF-C、IGF-Ⅰ高表达及SULT2A1低表达是子宫肌瘤患者发病的风险预测因子。 Objective To investigate the expression and correlation of sulfate-based transferase 2A1(SULT2A1),vascular endothelial growth factor-C(VEGF-C),and insulin-like growth factor-Ⅰ(IGF-Ⅰ)in patients with uterine fibroids.Methods A total of 50 cases of female uterine fibroids tissue and parasyoma tissue specimens preserved in the pathology department of Yixing People′s Hospital Affiliated to Jiangsu University from December 2020 to December 2022,and 65 cases of normal uterine muscle tissue preserved in the pathology department of the same hospital in the same period were selected as study project.The SULT2A1 and VEGF-C proteins were tested by immunohistochemistry,and the IGF-Ⅰexpression was tested by flow cytometry;the receiver operating characteristic(ROC)curve analysis of the predictive value of SULT2A1,VEGF-C and IGF-I alone and together for uterine fibroids;analysis of the correlation between SULT2A1,VEGF-C and IGF-Ⅰ.Results Compared with normal uterine muscle tissue and paratumoral tissue,uterine fibroid tissue showed higher VEGF-C,IGF-Ⅰexpression and lower SULT2A1 expression(P<0.05).The expression of VEGF-C and IGF-Ⅰwas higher than normal uterine muscle tissue,and SULT2A1 was lower than normal uterine muscle tissue(P<0.05).Correlation analysis showed that SULT2A1 and VEGF-C were negatively correlated(r=-0.902,P=0.001),IGF-Ⅰand VEGF-C were positively correlated(r=0.860,P=0.001),and SULT2A1 and IGF-Ⅰwere negatively correlated(r=-0.854,P=0.001);ROC showed that the predictive value of VEGF-C,IGF-Ⅰand SULT2A1 alone was lower than the three combinations.Conclusions The high expression of VEGF-C and IGF in uterine fibroids tissue and low expression of SULT2A1 in uterine fibroids tissue are correlated.The high expression of VEGF-C and IGF-Ⅰand low expression of SULT2A1 are predictors of risk in patients with uterine fibroids.
