运动训练配合穴位埋线对缺血性脑卒中大鼠运动功能的影响

The Influence of Exercise Training Combined with Catgut Embedment in Acupoint on Motor Function of Rats with Ischemic Stroke

摘要 目的:研究运动训练配合穴位埋线对脑缺血性脑卒中大鼠运动功能的影响,并探讨其作用机制。方法:将60只健康雄性SPF级SD大鼠随机分为假手术组、模型组、穴位埋线组、运动训练组和联合组,每组各12只。除假手术组外,其余组大鼠均制备缺血性脑卒中大鼠模型。造模完成后开始干预,穴位埋线组进行穴位埋线,运动训练组进行跑轮训练,联合组同时给予跑轮训练和穴位埋线治疗,连续14 d。假手术组和模型组不进行干预。干预的第1天、第7天和第14天,根据Garcia评分对大鼠的神经功能进行评分;采用网屏实验评价大鼠左侧肢体的前爪抓握能力及肌力变化;采用平衡木实验对大鼠平衡稳定性进行评估。苏木精-伊红(hematoxylin-eosin, HE)染色观察各组大鼠脑组织形态学变化。结果:与假手术组比较,造模大鼠的神经功能评分、网屏实验得分降低,平衡功能评分、平衡木实验到达时间和平衡木实验误足次数升高,差异均有统计学意义(P<0.05)。与模型组比较,第7天和第14天,运动训练组、穴位埋线组和联合组大鼠神经功能评分、网屏实验得分升高,平衡功能评分、平衡木实验到达时间和平衡木实验误足次数降低,且联合组的作用效果明显优于运动训练组和穴位埋线组,差异均有统计学意义(P<0.05)。HE染色显示,造模大鼠脑组织海马神经细胞排列紊乱,细胞结构不完整、有空泡状、神经元数量减少;其中联合组病理损害最轻。结论:运动训练联合穴位埋线可有效减轻缺血性脑卒中大鼠脑组织损伤,有效改善神经功能及运动功能障碍。 Objective:To study the influence of exercise training combined with catgut embedment in acupoint on motor function of rats with ischemic stroke and to explore its mechanism.Methods:A total of 60 healthy male SPF-grade SD rats were randomly divided into the sham operation group,the model group,the group of catgut embedment in acupoint,the exercise training group and the combined group,with 12 rats in each group.Except the sham operation group,all the other groups prepared ischemic stroke rats models.After the modeling was completed,the intervention began:the group of catgut embedment in acupoint was given catgut embedment in acupoint,the exercise training group was given wheel training,and the combined group was given wheel training and catgut embedment in acupoint for 14 consecutive days.The sham operation group and the model group did not receive any intervention.On the 1st,7th and 14th day of the intervention,the neurological function of rats was scored respectively according to the Garcia score.The changes in the grasping ability and muscle strength of the left limb of rats were evaluated by the net screen test.The balance stability of rats was evaluated by the bal-ance beam test.The morphological changes in brain tissues of rats in each group were observed by HE staining.Results:Compared with the sham operation group,the neurological function score and net screen test score of the model rats were decreased,while the balance function score,arrival time of balance beam test and the number of false steps in the balance beam test were increased,with statistically significant differences(P<0.05).Compared with the model group,on the 7th and 14th day,the neurological function score and net screen test score of the exercise training group,the group of catgut embedment in acupoint,and the combined group were increased,while the balance function score,arrival time of balance beam test and the number of false steps in the balance beam test were de-creased,and the effect of the combined group was significantly better than that of the exercise training group and the group of catgut em-bedment in acupoint,with statistically significant dfferences(P<0.05).HE staining showed that the brain tissue of the model rats was disordered,with incomplete cell structure,vacuolar,and reduced number of neurons.The pathological damage of the combined group was the least.Conclusion:Exercise training combined with catgut embedment in acupoint can effectively reduce ischemic brain tissue injury of the patients,improve neurological function,and significantly improve their motor dysfunction after stroke.
