Tigolaner及其类似物WY-403的合成与杀虫活性

Synthesis and insecticidal activity of tigolaner and its analogue WY-403

摘要 [目的]为探索tigolaner及其类似物WY-403在农业害虫防治领域的应用前景。[方法]以全氟-4-甲基-2-戊烯为原料,经过4步反应制得目标化合物,并测试其对鳞翅目害虫小菜蛾、半翅目害虫桃蚜及蜱螨目害螨朱砂叶螨的杀虫活性。以秀丽隐杆线虫谷氨酸门控性氯离子通道受体蛋白的晶体结构为模板构建了果蝇RDL受体蛋白模型,对其合理性进行了评价,并进行了分子对接。[结果]质量浓度为1.25 mg/L时,tigolaner及WY-403对朱砂叶螨和小菜蛾的致死率均可达到100%;质量浓度为10 mg/L时,tigolaner和WY-403对桃蚜的致死率分别高达100%和96.4%。分子对接结果表明:tigolaner及WY-403与受体蛋白之间存在氢键、疏水作用,同时还存在着较强的亲和力。[结论]Tigolaner和WY-403杀虫杀螨效果优异,在农业害虫防治领域具有广阔的应用前景。 [Aims]This study aims to explore the application prospects of tigolaner and its analogue WY-403 in the field of agricultural pest control.[Methods]The target compounds were synthesized through 4 steps with perfluoro(4-methylpent-2-ene)as the raw material,and the insecticidal activity against lepidopteran insect Plutella xylostella,hemipteran insect Myzus persicae,and acarid mite Tetranychus cinnabarinus were tested.Based on the crystal structure of glutamate-gated chloride channel receptor protein of Caenorhabditis ele gans,a model of the Drosophila RDL receptor protein was constructed,its rationality was evaluated by Ramachandran plot,and molecular docking was carried out.[Results]At a mass concentration of 1.25 mg/L,tigolaner and WY-403 could achieve 100%mortality against T.cinnabarinus and P.xylostella;at a mass concentration of 10 mg/L,tigolaner and WY-403 had 100%and 96.4%mortality against M.persicae,respectively.The molecular docking results showed that there were hydrogen bonds and hydrophobic interactions between target compounds and receptor protein,which indicated that there was a strong affinity between them.[Conclusions]Tigolaner and WY-403 have excellent insecticidal and acaricidal effects,and have broad application prospects in agricultural pest control.
