基于巨噬细胞极化探讨化浊解毒颗粒对大鼠急性肺损伤的影响

摘要 目的探讨化浊解毒颗粒对急性肺损伤大鼠巨噬细胞极化的影响。方法将60只SD雄性大鼠随机分为空白对照组、模型组、泼尼松组及化浊解毒低、中、高剂量组,每组10只。化浊解毒低、中、高剂量组分别给予14 g/kg、28 g/kg、56 g/kg的化浊解毒颗粒悬液灌胃,泼尼松组给予3 mg/kg醋酸泼尼松稀释液灌胃,空白对照组和模型组给予等体积蒸馏水灌胃,均1次/d,连续7 d。末次灌胃24 h后,除空白对照组外,其余各组大鼠采用气管滴注脂多糖2 mg/kg法制备急性肺损伤模型,造模3 h后取材。HE染色观察肺组织病理形态,流式细胞术检测支气管肺泡灌洗液和肺组织中CD86+M1型肺泡巨噬细胞、肺间质巨噬细胞和CD206+M2型肺泡巨噬细胞、肺间质巨噬细胞百分比。结果与空白对照组比较,模型组大鼠肺组织损伤严重,肺泡结构破坏明显,肺泡壁增厚,炎性细胞与红细胞浸润;CD86+M1型肺泡巨噬细胞、肺间质巨噬细胞百分比均显著升高(P均<0.05),CD206+M2型肺泡巨噬细胞、肺间质巨噬细胞百分比均显著降低(P均<0.05)。与模型组比较,各药物组大鼠肺组织损伤均减轻,CD86+M1型肺泡巨噬细胞、肺间质巨噬细胞百分比均显著降低(P均<0.05),CD206+M2型肺泡巨噬细胞、肺间质巨噬细胞百分比均显著升高(P均<0.05);化浊解毒各组CD86+M1型、CD206+M2型肺泡巨噬细胞、肺间质巨噬细胞百分比变化与给药剂量呈正相关。结论化浊解毒颗粒可通过调节巨噬细胞极化减轻大鼠急性肺损伤。 Objective It is to investigate the effect of Huazhuo Jiedu granule on macrophage polarization in rats with acute lung injury.Methods Sixty SD male rats were randomly divided into blank control group,model group,prednisone group,low,medium and high dose groups of Huazhuo Jiedu granule,with 10 rats in each group.The low,medium and high dose groups of Huazhuo Jiedu granule were intragastrically administered with 14 g/kg,28 g/kg and 56 g/kg of Huazhuo Jiedu granule suspension by gavage,the prednisone group was given 3 mg/kg of prednisone acetate dilution by gavage,and the blank control group and the model group were given an equal volume of distilled water by gavage,all once daily,continuously treated for 7 days.At 24 hours after the last gavage,the rats in all groups except for the blank control group were given lipopolysaccharide 2 mg/kg by tracheal drip to establish models of acute lung injury,and the examples were taken at 3h after modeling.The pathological morphology of lung histology was observed by HE staining,and the percentages of CD86+M1-type alveolar macrophage,lung interstitial macrophage,and CD206+M2-type alveolar macrophage,lung interstitial macrophage in bronchoalveolar lavage fluid and lung tissues were detected by flow cytometry.Results Compared with the blank control group,the lung tissue of rats in the model group was severely damaged,the alveolar structure was obviously destroyed,the alveolar wall was thickened,and inflammatory cells and red blood cells were infiltrated;the percentages of CD86+M1 alveolar macrophages and pulmonary interstitial macrophages were significantly increased(all P<0.05),and the percentages of CD206+M2 alveolar macrophages and pulmonary interstitial macrophages were significantly decreased(all P<0.05).Compared with the model group,the injury of lung tissue was alleviated in each group,the percentage of CD86+M1 alveolar macrophages and pulmonary interstitial macrophages was significantly decreased(all P<0.05),and the percentage of CD206+M2 alveolar macrophages and pulmonary interstitial macrophages was significantly increased(all P<0.05);the percentage of alveolar macrophages and pulmonary interstitial macrophages CD86+M1 and CD206+M2 cells in each group was positively correlated with the administered dose.Conclusion Huazhuo Jiedu granule can alleviate acute lung injury in rats by regulating macrophage polarization.
