蓝刺头对去卵巢大鼠血清P1NP、β-CTX及TNF-α、IL-1β、IL-6的影响

摘要 目的:研究蒙药蓝刺头对去卵巢大鼠骨代谢标志物P1NP、β-CTX的影响。方法:将72只4月龄SD系清洁级雌性大鼠随机分为六组,每组12只,分别为假手术组、去卵巢骨质疏松模型组、蓝刺头高剂量组、中剂量组、低剂量组及淫羊藿组。除假手术组外,其余各组大鼠均采用卵巢摘除去势法建立绝经后骨质疏松模型。术后正常饲养90 d,假手术组和模型组大鼠灌胃等体积0.9%氯化钠溶液,各治疗组大鼠灌胃相应药物,每天1次,连续90 d。末次给药24 h后,处死大鼠,采用Micro-CT测定大鼠股骨骨密度,酶联免疫法检测大鼠血清中P1NP、β-CTX、TNF-α、IL-1β、IL-6的含量。结果:与假手术组比较,模型组大鼠股骨近端BMD明显降低,血清中P1NP、β-CTX、TNF-α及IL-1β和IL-6的浓度明显升高(P<0.05)。与模型组比较,蓝刺头组的BMD升高,血清中P1NP、β-CTX、TNF-α及IL-1β和IL-6的浓度下降(P<0.05)。结论:蒙药蓝刺头可降低去卵巢大鼠的P1NP和β-CTX的含量,同时减少TNF-α、IL-1β、IL-6的浓度,进而达到增加骨含量,减少骨吸收的作用。 Objective:To study the effects of Eechinops latifolius on bone metabolic markers P1NP andβ-CTX in ovariectomized rats.Methods:Seventy-two 4-month-old SD SPF grade female rats were randomly divided into sham group,ovariectomized osteoporosis model group,Mongolian medicine Cyanostis high-dose group,medium-dose group,low-dose group and Epimedium group,12 rats in each group.Except sham operation group,the other groups were treated with ovary removal and castration to establish postmenopausal osteoporosis model.Rats in sham operation group and model group were fed normal saline for 90 days after operation,and rats in each treatment group were intragastric with corresponding drugs once a day for 90 days.24 h after the last administration,the rats were killed,and the femoral bone mineral density of the rats was determined by Micro-CT,and the serum levels of P1NP,β-CTX,TNF-α,IL-1βand IL-6 were detected by enzyme-linked immunoassay.Results:Compared with sham group,the proximal femur BMD of OVX group was significantly decreased,and the serum contents of P1NP,β-CTX,TNF-α,IL-1βand IL-6 were significantly increased(P<0.05).Compared with OVX group,BMD was increased and serum contents of P1NP,β-CTX,TNF-α,IL-1βand IL-6 were decreased in treatment group(P<0.05).Conclusion:Mongolian medicine Lanacitou can decrease the contents of P1NP andβ-CTX in ovariectomized rats,and at the same time decrease the concentrations of TNF-α,IL-1βand IL-6,thereby increasing bone content and decreasing bone absorption.
