瑶药黄钻总三萜超声辅助提取工艺优化及抗氧化、抑菌性研究

Optimization of ultrasound-assisted extraction of total triterpene from schisandra henryi clarke and study on its antioxidant and antibacterial properties

摘要 以瑶药黄钻为研究对象,通过响应面法优化黄钻中总三萜的超声提取工艺,并对三萜抗氧化能力及抑菌性进行测定。通过研究溶剂种类、溶剂浓度、料液比、超声时间对黄钻总三萜提取率的影响,利用Box-Behnken响应面法优化超声提取工艺。试验结果表明:溶剂甲醇浓度为72%、料液比为1:29、超声时间为41 min的提取条件下,总三萜提取率最优,为1.275%,与预测提取率基本一致,模型可靠。测定其总抗氧化能力,可知总三萜抗氧化能力良好;进行抑菌性测试,发现总三萜对大肠杆菌有抑制作用,最小抑菌浓度为25 mg/mL,对金黄色葡萄球菌无明显的抑制作用。研究为瑶药黄钻的开发利用提供参考。 With Schisandra henryi Clarke as the research subject,the ultrasonic extraction technology of total triterpenoids from Schisandra henryi Clarke was optimized by response surface method,and the antioxidant capacity and antibacterial activity of total triterpenoids were measured.By studying the effects of solvent type,solvent concentration,solid-liquid ratio and ultrasonic time on the extraction rate of total triterpene,Box-Behnken response surface method was used to optimize the ultrasonic extraction process.The experimental results show that under the conditions of methanol concentration of 72%,solid-liquid ratio of 1:29 and ultrasonic time of 41 min,the extraction rate of total triterpene is optimal,which is 1.275%,which is basically consistent with the predicted extraction rate.The model is reliable.The total antioxidant capacity of triterpenoids was determined.It is known that total triterpenes have good antioxidant capacity.Through bacteriostatic testing,it is found that the minimum inhibitory concentration of total triterpenes on E.coli is 25 mg/mL,and the inhibitory effect of total triterpenes on Staphylococcus aureus at this concentration is not obvious.The study provides a reference for the development and utilization of Schisandra henryi clarke.
