Notch信号通路在曲安奈德治疗裸鼠血管瘤模型过程中的表达

摘要 目的探究细胞增殖分化的Notch信号通路在曲安奈德治疗裸鼠血管瘤模型过程中的表达情况及其作用机制。方法选取2022年12月—2023年6月深圳市龙岗中心医院18只小鼠。分为Control组、Control+TA for 7 days处理组、Control+TA for 14 days处理组,每组6只,并进行相应给药处理,每3 d记录肿瘤生长状况,利用蛋白质免疫印迹(western blot,WB)和实时荧光定量聚合酶链式反应(real-time quantitative polymerase chain reaction,qPCR)检测糖皮质激素受体α(glucocorticoid receptorα,GRα)、Notch1、表皮生长因子受体(epidermal growth factor receptor,EGRF)和Split多毛增强子(hairy and enhancer of split,HES1)蛋白表达情况,免疫组化观察阳性细胞。结果WB实验结果显示,与Control组相比,Control+TA for 7 days处理组和Control+TA for 14 days处理组中GRα、Notch1和EGFR蛋白表达显著下降,而HES1蛋白显著上升,差异有统计学意义(P<0.05);qPCR实验结果显示,与对照组相比,Control+TA for 7 days处理组和Control+TA for 14 days处理组中GRα、Notch1和HES1蛋白表达显著下降,而EGFR蛋白显著上升,差异有统计学意义(P<0.05);免疫组化结果显示,与Control组相比,Control+TA for 7 days处理组免疫组化和Control+TA for 14 days处理组中GRα、Notch1、HES1和EGFR阳性细胞减少,差异有统计学意义(P<0.05)。结论曲安奈德可使裸鼠模型血管瘤组织GRα、Notch1表达下降,Notch信号通路参与曲安奈德治疗血管瘤的作用过程。 Objective To investigate the expression of Notch signaling pathway of cell proliferation and differentiation during triamcinolone acetonide treatment of nude mouse hemangioma model,and to explore its mechanism of action.Methods 18 mice were selected from December 2022 to June 2023 in Longgang Central Hospital.They were divided into control group,control+TA for 7 days treatment group,control+TA for 14 days treatment group,6 mice in each group,and were given drug treatment accordingly,tumor growth was recorded every 3 days,glucocorticoid receptorα(GRα),Notch1,epidermal growth factor receptor(EGFR)and hairy and enhancer of split(HES1)protein expression was detected by western blot(WB)and real-time quantitative polymerase chain reaction(qPCR),and immunohistochemistry was performed to observe the positive cells.Results The results of WB experiments showed that compared with the control group,the expression of GRα,Notch1 and EGFR proteins decreased significantly in the control+TA for 7 days treatment group and the control+TA for 14 days treatment group,while the HES1 protein increased,and the differences were statistically significant(P<0.05).The results of qPCR experiments showed that the expression of GRα,Notch1 and HES1 proteins was significantly decreased,while EGFR protein was significantly increased in control+TA for 7 days treatment group and control+TA for 14 days treatment group compared with control group,and the differences were statistically significant(P<0.05).Immunohistochemistry results showed a decrease in GRα,Notch1,HES1,and EGFR-positive cells in the control+TA for 7 days treatment group immunohistochemistry and control+TA for 14 days treatment group compared with the control group,and the differences were statistically significant(P<0.05).Conclusion Triamcinolone acetonide could decrease the expression of GRαand Notch1 in hemangioma tissues of nude mice model.Notch signaling pathway is involved in the course of action of triamcinolone acetonide in the treatment of hemangioma.
