摘要
对蓖麻叶中提取的总黄酮进行纯化及抗氧化活性研究。通过比较6种大孔树脂的吸附和解吸能力,最终确定HPD100型为最优。结果表明:上样浓度为0.98 mg·mL^(-1)、上样流速为2 BV·h^(-1)、pH为4、洗脱剂70%乙醇、洗脱流速为3 BV·h^(-1)时纯化效果最好,蓖麻总黄酮纯度达到56.34%;纯化后的蓖麻总黄酮质量浓度为1.0 mg·mL^(-1)时对DPPH的清除率为78.51%;HPD100大孔树脂对蓖麻总黄酮纯化效果好,蓖麻总黄酮具有一定抗氧化活性。
The purification conditions and antioxidant activity of total flavonoids from castor leaves were studied in this paper.By comparing the adsorption and desorption capacity of six macroporous resins,HPD100 was final⁃ly determined to be the best.The results showed that the optimal purification conditions were as follows:the con⁃centration of loading sample was 0.98 mg·mL^(-1),and the flow rate was 2 BV·h^(-1),pH 4,and the extract concentration was 70%,and the elution flow rate was 3 BV·h^(-1).The purity of total flavonoids from castor leaves reached 56.34%.Total flavonoids at 1.0 mg·mL^(-1) demonstrated DPPH scavenging rate of 78.51%.It is concluded that HPD100 macroporous resin is suitable for the purification of total flavonoids from castor leaves and the total fla⁃vonoids have antioxidant activity.
