香菇柄多酚负压提取、纯化及体外活性评价

摘要 为提高香菇柄利用率,通过单因素和正交试验对香菇柄中多酚负压提取工艺进行优化,考察不同溶剂萃取香菇柄多酚粗提物组分的抗氧化活性,评价纯化对多酚降血糖活性的影响。结果表明,香菇柄多酚最佳负压提取工艺条件为真空度0.098 MPa、乙醇浓度40%、料液比1∶35(g/mL),浸提温度50℃,浸提时间50 min,此条件下香菇柄多酚提取量为3.07 mg/g。香菇柄多酚粗提物经石油醚、乙酸乙酯和正丁醇分别萃取后,正丁醇萃取组分多酚得率最高7.61%,乙酸乙酯萃取组分多酚含量最高64.17%,经乙酸乙酯萃取后单位质量浓度多酚对自由基的清除能力显著提升。体外降血糖结果显示,纯化前后香菇柄多酚对α-淀粉酶和α-葡萄糖苷酶均具有剂量依赖性抑制活性,表明香菇柄多酚具有一定的降血糖活性,纯化后多酚可显著提高对两种酶的抑制活性,表明纯化可提高多酚的降血糖活性。 In order to improve the utilization rate of Lentinus edodes stipe,the vacuum extraction process of polyphenols from L.edodes stipe was optimized by single factor and orthogonal test.The crude polyphenol extract from L.edodes stipe was extracted with different solvents,and the antioxidant activities of different extracted components were investigated.The extracted components were subjected to column chromatography to evaluate the effect of purification on hypoglycemic activity of polyphenols.The results showed that the optimum extraction conditions of polyphenol from L.edodes stipe under vacuum condition were as follows:vacuum degree 0.098 MPa,ethanol concentration 40%,solid-liquid ratio 1∶35(g/mL),extraction temperature 50℃and extraction time 50 min.Under these conditions,the yield of polyphenol from L.edodes stipe was 3.07 mg/g.When the crude extract of L.edodes stipe polyphenol was extracted by petroleum ether,ethyl acetate and n-butanol respectively,the polyphenol yield of n-butanol extraction component was the highest 7.61%,and the polyphenol content of ethyl acetate extraction component was the highest 64.17%,and the scavenging ability of polyphenol was significantly improved after extraction by ethyl acetate.The results of hypoglycemic activities in vitro showed that the both purified and not purified polyphenols of L.edodes stipe had dose-dependent inhibitory activities onα-amylase andα-glucosidase,which indicated that the polyphenols extract from L.edodes stipe had certain hypoglycemic activity,and the purified polyphenol could significantly improve the inhibitory activities of the two enzymes above,which indicated that purification could improve the hypoglycemic activity of polyphenol.
