摘要
本研究通过体外抗氧化实验和细胞损伤模型实验,研究蓝莓中花青素抗氧化能力。采用ABTS自由基清除实验、DPPH自由基清除实验、铁离子螯合能力实验和羟基自由基清除能力实验对100~500μg/mL不同浓度的花青素进行体外抗氧化活性评价。在氧化应激细胞模型实验中,通过测定H_(2)O_(2)诱导HepG2细胞中MDA含量、SOD活力、GSH含量探究100~500μg/mL不同浓度花青素对HepG2细胞氧化损伤的保护作用。结果表明,随着花青素浓度的增加,ABTS自由基清除能力、DPPH自由基清除能力、铁离子螯合能力和羟基自由基清除能力均随之增加,且比模型组具有显著的抗氧化效果(P<0.05)。花青素可减少H_(2)O_(2)诱导HepG2细胞氧化损伤时MDA含量,提高HepG2细胞内SOD活力及GSH含量,减少氧化损伤后细胞凋亡情况的发生,对HepG2细胞氧化损伤有一定保护作用。蓝莓中花青素可用于开发功能性食品以及作为食品添加剂用作抗氧化的良好材料,本研究可为花青素资源的开发利用提供理论基础。
The antioxidant capacity of anthocyanins in blueberry was studied by in vitro antioxidant experiment and cell damage model.The antioxidant activity of blueberry anthocyanins with different concentrations of 100~500μg/mL was evaluated by ABTS free radical scavenging,DPPH free radical scavenging,Fe^(2+)chelating ability and hydroxyl free radical scavenging ability.In the oxidative stress cell model,the protective effects of blueberry anthocyanins at different concentrations of 100~500μg/mL on oxidative damage of HepG2 cells were investigated by measuring MDA content,SOD activity and GSH content induced by H_(2)O_(2).The results showed that ABTS free radical scavenging ability,DPPH free radical scavenging ability,iron chelating ability and hydroxyl free radical scavenging ability were increased with the increase of blueberry anthocyanin concentration,and had a significant antioxidant effect compared with the model group(P<0.05).Blueberry anthocyanins could reduce MDA content in oxidative damage in HepG2 cells induced by H_(2)O_(2),increase SOD activity and GSH content in HepG2 cells,reduce cell apoptosis after oxidative damage,and have a certain protective effect on oxidative damage in HepG2 cells.The anthocyanins in blueberry could be used as good materials for developing functional food and additives for antioxidant.This study provides a theoretical basis for the development and utilization of anthocyanins resources.
