摘要
为揭示花期糯红高粱内生酵母群落结构及功能特点。利用纯培养法对宜宾地区酿酒专用糯红高粱花期内生酵母多样性及群落结构进行探究与分析,并通过选择培养基筛选其中的功能菌。结果表明,从花期糯红高粱中分离得到54株内生酵母菌菌株,鉴定为12个属17个种,优势属为隐球菌属(Cryptococcus),优势种为黄色隐球酵母(Cryptococcus luteolus)。糯红高粱花期根、茎、叶、花各组织中的内生酵母各项多样性指数均有明显差异,其丰富度指数表现为茎>根>花>叶;均匀度指数表现为根>花>叶>茎;优势度指数表现为花>根>茎>叶。花期糯红高粱内生酵母功能丰富,菌株FBJ15产淀粉酶能力最强,菌株FAF8产蛋白酶能力最强,菌株FAJ43产脂肪酶能力最强;HBF22菌株产酯酶能力最强。由此可知,糯红高粱花期内生酵母种类丰富且功能多样,该研究结果为深入探究内生酵母与宿主糯红高粱的互作机制提供参考。
To reveal the structure and functional characteristics of endophytic yeast community in waxy sorghum at flowering stage,the diversity and community structure of endophytic yeasts in waxy sorghum for brewing in Yibin region at flowering stage were analyzed and investigated using pure culture method,and the functional bacteria were screened by selecting medium.The results showed that 54 endophytic yeast strains were isolated from waxy sorghum at flowering stage,identified as 12 genera and 17 species,and the dominant genus and species was Cryptococcus and Cryptococcus luteolus,respectively.The diversity index of endophytic yeasts in root,stem,leaf and flower tissues of waxy sorghum at flowering stage had obvious difference,the richness index was stem>root>flower>leaf,the evenness index was root>flower>leaf>stem,and the dominance degree index was flower>root>stem>leaf.The function of endophytic yeasts in waxy sorghum during flowering period was rich.Among them,the strain FBJ15had the strongest amylase-producing ability,the strain FAF8 had the strongest protease-producing ability,the strain FAJ43 had the strongest lipaseproducing ability,and the strain HBF22 had the strongest esterase-producing ability.Therefore,the endophytic yeasts in waxy sorghum at flowering stage were rich in types and diverse in functions.These results could provide a reference for further exploring the interaction mechanism between endophytic yeast and host waxy sorghum.
