基于杜仲降压片指纹图谱的化学模式识别及对松脂醇二葡萄糖苷的含量测定

摘要 目的结合成分定量与化学模式识别建立杜仲降压片的HPLC指纹图谱,评价杜仲降压片的差异,并且测定松脂醇二葡萄糖苷含量。方法采用色谱柱Ultimate AQ-C_(18)(250 mm×4.6 mm,5μm),以乙腈-水为流动相进行梯度洗脱,检测波长为277 nm,对10批杜仲降压片样品进行指纹图谱研究,利用化学模式识别法对共有峰面积色谱数据进行聚类分析和主成分分析,对不同批次的杜仲降压片样品进行质量评价,并对其松脂醇二葡萄糖苷进行含量测定。结果建立了杜仲降压片指纹图谱,包含15个共有峰。并对共有峰的面积作为变量进行了化学模式识别,可将不同公司的杜仲降压片大致分为两大类。松脂醇二葡萄糖苷在0.0625~1.0000 mg/mL内线性关系良好;平均加样回收率为111.24%,RSD为1.69%。结论通过指纹图谱结合化学模式识别技术评价杜仲降压片的差异,该方简便可行且重复性强,以期为杜仲降压片的质量控制提供了依据。 Objective To establish the HPLC fingerprint of Eucommia Antihypertensive Tablets,evaluate the differences of Eucommia Antihypertensive Tablets by combining component quantification and chemical pattern recognition,and determine the content of pinethyl alcohol diaglucoside.Methods Column Ultimate AQ-C_(18)(250 mm×4.6 mm,5 m)was used to elute gradient with acetonitrile-water as the mobile phase at a detection wavelength of 277 nm,10 batches of Eucommia decompression sheet samples were fingerprinted,cluster analysis and principal component analysis of common peak area chromatography data were performed by chemical pattern recognition,the quality of different batches of Eucommia Antihypertensive Tablets samples was evaluated,and the content of pinethyl alcohol diaglucoside was determined.Results The fingerprint of Eucommia Antihypertensive Tablets film was established,containing 15 common peaks.The area of the common peak was identified as a variable,and the Eucommia Antihypertensive Tablets of different companies could be roughly divided into two categories.Moreover,the linear relationship between pinitol discoside in 0.0625~1.0000 mg/mL was good.The average spiked recovery rate was 111.24%and RSD was 1.69%.Conclusion The evaluation of the differences of Eucommia Antihypertensive Tablets by fingerprinting combined with chemical pattern recognition technology is simple,feasible and reproducible,providing a basis for the quality control of Eucommia Antihypertensive Tablets.
