摘要
探索黄精多糖最佳脱蛋白方法;以蛋白脱除率及多糖保留率为指标考察三氯醋酸(TCA)法、Sevage试剂法、盐析法、盐酸法对黄精多糖脱蛋白影响,通过不同方法脱蛋白黄精多糖对DPPH自由基、O_(2)^(-)自由基清除率评价其体外抗氧化能力。黄精多糖蛋白脱除率为Sevage试剂法>TCA法>盐酸法>盐析法;多糖保留率为TCA法>盐酸法>Sevage试剂法>盐析法,体外抗氧化活性为TCA法>盐酸法>Sevage试剂法>盐析法。不同脱蛋白方法对黄精多糖蛋白脱除率及多糖保留率及其体外抗氧化活性产生不同的影响,其TCA法是一种较好的黄精多糖脱蛋白的方法。
To explore the best deproteinization method of Polygonatum sibiricum polysaccharide.The trichloroacetic acid(TCA)method,Sevage method,salting-out method and hydrochloric acid method on the deproteinization of polyvitriol polysaccharide were investigated with the removal rate of protein and polysaccharide retention rate as index,and their antioxidant activity wasdetermined by measuring the scavenging rate of DPPH radicals,O_(2)^(-)radicals.The results showed that the removal rate of protein were Sevage method>TCA method>hydrochloric acid method>salting-out method.The polysaccharide retention rate was TCA method>hydrochloric acid method>Sevage method>salting-out method,and in vitro antioxidant activity was TCA method>hydrochloric acid method>Sevage method>salting-out method.Different deprotein-ization methods have different effects on the removal rate of protein,polysaccharide retention rate and its antioxidant activity in vitro,among which TCA method is a better method.
