长林小蠹肠道可培养细菌群落的组成和结构

摘要 为了研究长林小蠹(Hylurgus ligniperda Fabricius)肠道可培养细菌群落的组成和结构。采用传统细菌分离培养法对采集自山东省烟台市牟平区的长林小蠹成虫与幼虫的肠道细菌进行分离纯化,对分离获得的细菌结合形态学及16S rDNA测序方法进行细菌种类鉴定,初步判定其分类学地位。从长林小蠹成虫和幼虫肠道中分离鉴定出了20株细菌,它们隶属于2门4纲6目9科15属18种,优势菌门为变形菌门(Proteobacteria)(91.9%),优势菌属为沙雷氏菌属(Serratia)(27.7%)和Rouxiella属(14.8%),优势种为黏质沙雷氏菌Serratia marcescens(27.7%)和Rouxiella silvae(14.8%)。主要包括,芽孢杆菌属(Bacillus)1种;乳球菌属(Lactococcus)1种;布鲁氏菌(Brucella)1种;帕拉伯克霍尔德氏属(Paraburkholderia)1种;伯克霍尔德氏属(Burkholderia)1种;泛菌属(Pantoea)2种;欧文氏菌属(Erwinia)2种;沙雷氏菌属(Serratia)1种;(Rouxiella)属1种;科萨克氏菌属(Kosakonia)1种;克雷伯氏菌属(Klebsiella)1种;拉乌尔菌属(Raoultella)2种;Atlantibacter属1种;哈夫尼亚菌属(Hafnia)1种;假单胞菌属(Pseudomonas)1种。其中,布鲁氏菌属和Atlantibacter属是长林小蠹区别于目前我国分布的其他小蠹虫肠道可培养细菌所特有的属。 In order to study the composition and structure of the intestinal culturable bacterial community of Hylurgus ligniperda Fabricius.Traditional bacterial isolation and cultivation methods were used to isolate and purify intestinal bacteria from adult and larval H.ligniperda collected from Mouping District,Yantai City,Shandong Province.The morphological method combined with 16S rDNA sequencing was used to identify the species of the isolated bacteria and preliminarily determine their taxonomic status.A total of 20 bacterial strains were isolated and identified from the gut of adult and larval H.ligniperda,belonging to 2 phyla,4 classes,6 orders,9 families,15 genera,and 18 species.The dominant bacteriophyta bacterium was Proteobacteria(91.9%),and the dominant bacterium was Serratia(27.7%)and Rouxiella(14.8%).The dominant species was Serratia marcescens(27.7%)and Rouxiella silvae(14.8%).Mainly includes 1 species of Bacillus,1 species of Lactococcus,1 species of Brucella,1 species of Paraburkholderia,1 species of Burkholderia,2 species of Pantoea,2 species of Erwinia,1 species of Serratia,1 species of Rouxiella,1 species of Kosakonia,1 species of Klebsiella,2 species of Raoultella,1 species of Atlantibacter,1 species of Hafnia,1 species of Pseudomonas.Among them,the genera Brucella and Atlantibacter are unique to the culturable bacteria in the intestinal tracts of Hylurgus distributed in our country.
