摘要 目的:优化远志皂苷类成分的提取工艺,方法:以皂苷含量为指标,进行单因素考,包括提取方法、提取次数、提取溶剂浓度、料液比和提取时间,确定对远志中皂苷含量影响较大的因素。采用box-behnken(BBD)响应面法优化远志皂苷提取工艺。结果表明:远志皂苷类成分的最佳提取工艺参数为乙醇浓度80%、提取时间1.5/h、料液比1∶10 g/mL,在此优化条件下皂苷含量5.48 mg/mL,与预测值5.45 mg/mL,有良好的拟合性,证实此工艺可行,预测性良好。 Objective:To optimize the extraction process of saponin components in Polygala tenuifolia.Method:Using saponin content as an indicator,a single factor analysis was conducted,including extraction method,extraction solvent,extraction frequency,ethanol concentration,solid-liquid ratio,and extraction time,to determine the factors that have a significant impact on the saponin content in Polygala tenuifolia.The box Behnken(BBD)response surface methodology was used to optimize the extraction process of saponins from Radix Polygalae.The results showed that the optimal extraction process parameters for saponin components in Polygala tenuifolia were ethanol concentration of 80%,extraction time of 1.5 hours,and solid-liquid ratio of 1:10 g/mL.Under these optimized conditions,the saponin content was 5.48 mg/mL,which had a good fit with the predicted value of 5.45 mg/mL,confirming the feasibility of this process and good predictability.
