PD-1、PD-L1在子痫前期孕妇外周血及胎盘组织中的表达及意义

摘要 目的分析子痫前期患者外周血及胎盘组织中程序性死亡受体-1(PD-1)、PD-配体1(PD-L1)的表达及其意义。方法用流式细胞法和免疫组化法测定子痫前期孕妇(观察组,n=30)和正常孕妇(对照组,n=30)外周血T淋巴细胞表面及胎盘组织中的PD-1、PD-L1表达,并分析其与孕妇平均动脉压、24 h尿蛋白等指标的相关性。结果观察组孕妇外周血T细胞表面及胎盘组织中的PD-1、PD-L1表达水平均明显低于对照组(P<0.05),PD-1、PD-L1在外周血的表达水平与胎盘组织中的表达呈正相关(r=0.432、0.377,均P<0.05)。PD-1、PD-L1水平与孕妇平均动脉压、24 h尿蛋白呈负相关(P<0.05)。结论PD-1、PD-L1在子痫前期孕妇外周血的表达可反映其在胎盘组织中的表达,且其表达水平可反映病情的严重程度。 Objective To analyze the expression of PD-1 and PD-L1 in peripheral blood and placental tissues of patients with preeclampsia and their significance.Methods The expressions of PD-1 and PD-L1 on the surface of peripheral blood T lymphocytes and in placental tissues in 30 preeclampsia pregnant women(Observation Group,n=30)and 30 normal pregnant women(Control Group,n=30)were determined by flow cytometry and immunohistochemistry,and their correlation with the mean arterial pressure,24 h urine protein and other indicators of the pregnant women were analyzed.Results The expressions of PD-1 and PD-L1 on the surface of peripheral blood T lymphocytes and in placental tissues in the Observation Group were significantly lower than those in the Control Group(P<0.05).The expressions of PD-1 and PD-L1 in peripheral blood were positively correlated with those in placental tissue(r=0.432 and 0.377,P<0.05 for all).The expressions of PD-1 and PD-L1 were negatively correlated with mean arterial pressure and 24-hour urine protein of the pregnant women(P<0.05).Conclusion The expressions of PD-1 and PD-L1 in the peripheral blood of pregnant women with preeclampsia can reflect the expressions of PD-1 and PD-L1 in placenta tissues,and also reflect the severity of preeclampsia.
