摘要
数字PCR作为第三代PCR检测技术,无需校准物和绘制标准曲线即可实现对样品初始浓度的绝对定量,具有高灵敏度、高特异性和高精确度的特点;已被广泛应用于遗传病、癌症、传染病的诊断和拷贝数变异、突变检测、转基因检测等方面的研究。本文对数字PCR技术的发展、原理、数据分析做了介绍,并对数字PCR技术在国门生物安全领域的研究进展和应用前景做了介绍和论述。
As the third generation PCR detection technology,digital PCR can absolutely quantify the initial concentration of nucleic acid in samples without calibrators and standard curves,which has the characteristics of high sensitivity,high specificity and high accuracy.It has been widely used in the diagnosis of genetic diseases,cancer,infectious diseases,and the study of copy number variation,mutation detection,transgene detection and other aspects.In this paper,the development,principle and data analysis methods of digital PCR technology are introduced,the research progress and application prospect of digital PCR technology in the field of customs bio-security are introduced and discussed.
