番茄SlFHA1基因的克隆及生物学信息分析

摘要 为明确番茄SlFHA1基因的基本性质及其在盐胁迫下的表达特征,从番茄品种“895”叶片中成功克隆出SlFHA1基因,并对其进行生物信息学分析,在番茄中各器官中和不同盐胁迫下的表达分析,为研究番茄的耐盐分子机制提供理论依据。结果表明,SlFHA1的cDNA序列全长为1023 bp,编340个氨基酸;SlFHA1的蛋白相对分子质量为37613.47,分子式为C1671H2585N469O499S12,理论等电点为6.40,为疏水性蛋白,且是没有信号肽和跨膜结构的蛋白。SlFHA1蛋白内含FHA结构域,属于forkhead family超家族基因,定位于细胞核中。SlFHA1在番茄的根、茎、叶中均有表达,且在叶中表达水平最高;盐处理后,SlFHA1的表达量升高。 We successfully cloned the SlFHA1 gene from the leaves of tomato cultivar"895",and analyzed its bioinformatics and expression in various organs in tomato under different salt stress,this provides the theoretical basis for the molecular mechanism of salt tolerance in tomato.The results showed the cDNA sequence of SlFHA1 was 1023 bp and 340 amino acids.The relative molecular weight of SlFHA1 is 37613.47,the molecular formula is C1671H2585N469O499S12,and thetheoretical isoelectric point is 6.40.SlFHA1 protein is a hydrophobic protein with no signal peptide and no transmembranestructure and it contains FHA domain,belonging to the forkhead family superfamily gene localized in the nucleus.SlFHA1expresses in the roots,stems and leaves of tomatoes with the highest level in the leaves.The expression of SlFHA1is increased after salt treatment.
