高效液相色谱法测定保心宁胶囊中7个指标性成分的含量

Determination of Seven Index Components in Baoxinning Capsules by HPLC

目的 建立高效液相色谱法(HPLC)同时测定保心宁胶囊中丹参素、柚皮苷、橙皮苷、新橙皮苷、迷迭香酸、丹酚酸B和丹参酮II_A7个指标性成分的含量。方法 以Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm,5μm)为色谱柱,以乙腈(A)-0.1%磷酸(B)为流动相进行梯度洗脱;检测波长为283,270 nm;流速:1.0 ml/min,柱温:30℃。结果 丹参素、柚皮苷、橙皮苷、新橙皮苷、迷迭香酸、丹酚酸B和丹参酮II_A在各自的测定范围内线性关系良好,相关系数(r)均大于0.999,平均回收率在97.62%~99.28%之间,RSD均小于2.0%。9批次保心宁胶囊中丹参素、柚皮苷、橙皮苷、新橙皮苷、迷迭香酸、丹酚酸B、丹参酮Ⅱ_A含量分别为1.289~1.506,10.337~12.083,0.883~1.136,8.687~10.103,0.876~1.031,7.494~9.001,0.971~1.403 mg/粒。结论 所建立的测定方法简便、准确、灵敏、重复性较好,可用于保心宁胶囊的质量控制。

Objective To establish an HPLC method for simultaneous determination of seven index components including salvianic acid A,naringin,hesperidin,neohesperidin,rosmarinic acid,salvianolic acid B and tanshinone IIA in Baoxinning Capsules.Methods An Agilent ZORBAX SB-C18(4.6 mm×250 mm,5μm)column was adopted,using acetonitrile(A)-0.1%phosphoric acid(B)as mobile phase for gradient elution.The detection wavelengths were set at 283 nm and 270 nm,the flow rate was 1.0 ml/min,and the column temperature was 30℃.Results The linear relationships of salvianic acid A,naringin,hesperidin,neohesperidin,rosmarinic acid,salvianolic acid B and tanshinone IIA were good within their respective determination ranges.The correlation coefficients(r)were greater than 0.999,the average recoveries were 97.62%-99.28%,with RSDs less than 2.0%.The contents of salvianic acid A,naringin,hesperidin,neohesperidin,rosmarinic acid,salvianolic acid B and tanshinone IIA in nine batches of Baoxinning Capsules were 1.289-1.506,10.337-12.083,0.883-1.136,8.687-10.103,0.876-1.031,7.494-9.001 and 0.971-1.403 mg per capsule.Conclusion The established method is simple,accurate,sensitive and reproducible,and can be used for the quality control of seven index components in Baoxinning Capsules.
