超声辅助高凝固点低共熔溶剂提取黄芪黄酮

摘要 为将新型绿色溶剂应用于传统中草药活性成分的提取,同时改善低共熔溶剂在提取领域的限制因素,本试验使用超声辅助低共熔溶剂提取黄芪中的五种黄酮类化合物。共合成了10种不同的低共熔溶剂,单因素试验考察了超声温度、超声时间、液固比、含水量和超声功率对五种黄酮得率的影响。在单因素试验的基础上,以五种黄酮的总得率为指标使用响应面优化法优化提取工艺。结果表明:低共熔溶剂氯化胆碱-草酸(摩尔比1:2)为最佳提取溶剂,优化后的提取条件为:超声时间43 min,超声温度44℃,含水量54%,在此条件下五种黄酮的总得率为1.70 mg/g,高于传统的甲醇提取法。因此,超声辅助低共熔溶剂提取法可有效提高黄芪黄酮的得率。 In order to apply the new green solvent to the extraction of active ingredients of traditional Chinese medicine and improve the limiting factors of deep eutectic solvent in the extraction field,ultrasound-assisted deep eutectic solvent was used to extract five flavonoids in Astragalus membranaceus.The best one for the extraction of five flavonoids from Astragalus membranaceus was selected from ten different deep eutectic solvents.The effects of ultrasonic temperature,ultrasonic time,liquid-solid ratio,water content and ultrasonic power on the yield of five flavonoids were investigated by single factor experiment.The results showed that the deep eutectic solvents choline chloride-oxalic acid(1:2)was the best extraction solvent.The optimal conditions were:ultrasonic time 43 min,ultrasonic temperature 44℃,and water content 54%.In this scenario,the total yield of five flavonoids reached 1.70 mg/g,which was higher than those obtained using conventional organic solvents.Therefore,ultrasonic-assisted deep eutectic solvents extraction could effectively improve the yield of Astragalus flavonoids.
