大孔树脂吸附法纯化酸枣仁总黄酮的工艺研究

摘要 利用7种大孔树脂对酸枣仁的总黄酮进行纯化,依据其吸附能力及解吸能力,选出最佳的大孔树脂型号,研究了上样液浓度、上样速度、上样液体积对大孔树脂吸附率的影响以及洗脱剂类型、洗脱剂浓度、洗脱速度、洗脱剂体积对大孔树脂解吸率的影响,采用正交试验对酸枣仁总黄酮的纯化工艺进行了优化。试验结果表明,DM301大孔树脂纯化酸枣仁总黄酮效果最佳,在上样液浓度为0.1 mg/mL、上样速度为1 BV/h、上样液体积为30 mL、洗脱剂为丙酮、洗脱剂浓度为100%、洗脱速度为2 BV/h、洗脱剂体积为55 mL的条件下,获得的酸枣仁总黄酮纯度最高,相较于纯化前提高了约1.1倍。 Seven kinds of macroporous resins are used to purify the total flavonoids from Ziziphi Spinosae Semen.According to the adsorption and desorption capacity,the best macroporous resin model is selected.The effects of loading solution concentration,loading speed and loading solution volume on the adsorption rate of macroporous resins and the effects of eluent type,eluent concentration,elution speed and eluent volume on the desorption rate of macroporous resin are studied.The purification process of total flavonoids from Ziziphi Spinosae Semen by macroporous resin is optimized by orthogonal test.The results show that DM301 macroporous resin has the best purification effect on the total flavonoids from Ziziphi Spinosae Semen.Under the conditions of loading solution concentration of 0.1 mg/mL,loading speed of 1 BV/h,loading solution volume of 30 mL,eluent of acetone,eluent concentration of 100%,elution speed of 2 BV/h and eluent volume of 55 mL,the purity of the total flavonoids from Ziziphi Spinosae Semen is the highest,which is 1.1 times higher than that before purification.
