染色质调控因子相关基因在乳癌预后评估中的价值

VALUE OF CHROMATIN REGULATOR-RELATED GENES IN EVALUATING THE PROGNOSIS OF BREAST CANCER

摘要 目的通过构建染色质调控因子(CRs)相关基因预后模型,探讨CRs相关基因在乳癌预后评估中的价值。方法在基因表达综合(GEO)数据库中进行差异性分析,筛选出CRs相关的差异基因。采用单因素Cox回归分析和LASSO回归方法建立CRs相关基因预后模型并进行验证。选取30对临床乳癌及癌旁组织样本,采用实时荧光定量聚合酶链反应(RT-qPCR)方法对关键CRs相关基因的表达进行临床验证。结果依据差异性分析结果建立基于4个CRs相关基因(MORF4L1、NCOA4、TTK和JMJD4)的乳癌病人预后模型,并根据模型风险评分临界值将病人分为高风险组和低风险组,生存分析显示高风险组预后较差。RT-qPCR结果显示,在乳癌组织中MORF4L1、NCOA4的表达下降,TTK、JMJD4的表达升高,与差异性分析的结果相一致。结论基于上述4个CRs相关基因的预后模型可以有效预测乳癌病人的预后。 Objective To construct a prognostic model of chromatin regulator(CR)-related genes,and to investigate the value of CR-related genes in evaluating the prognosis of breast cancer.Methods A difference analysis was performed in the Gene Expression Omnibus database to screen for the differentially expressed CR-related genes.Univariate Cox regression analysis and LASSO regression analysis were used to establish and validate the prognostic model based on CR-related genes.A total of 30 pairs of breast cancer tissue samples and adjacent tissue samples were selected,and RT-qPCR was used to verify the expression of key CR-related genes.Results According to the results of the difference analysis,a prognostic model of breast cancer patients was established based on four CR-related genes(MORF4L1,NCOA4,TTK,and JMJD4),and the patients were divided into high-and low-risk groups according to the cut-off value of the risk model.Survival analysis showed that the high-risk group tended to have a poor prognosis.The results of RT-qPCR showed that there were reductions in the expression of MORF4L1 and NCOA4 and increases in the expression of TTK and JMJD4 in breast cancer tissue,which was consistent with the results of the difference analysis.Conclusion The prognostic model based on the above four CR-related genes can effectively predict the prognosis of breast cancer patients.
