摘要
目的 探究纳米二氧化铈(CeO_(2)NPs)亚急性腹腔染毒对ICR小鼠体重、主要脏器以及血清氧化还原指标的影响。方法 将雄性ICR小鼠35只随机分成5组,每组7只。设生理盐水对照组和不同浓度的CeO_(2)NPs(100、500、1000、5000μg/kg)试验组。小鼠腹腔隔日注射CeO_(2)NPs, 28 d后进行相关指标的检测。结果 与对照组相比,CeO_(2)NPs各剂量组小鼠体重增长差异不显著(P>0.05);5000μg/kg剂量组肝系数、附睾系数与精子存活率与对照组相比差异极显著(P<0.01)。5000μg/kg剂量组肝和脾内可见纳米氧化铈沉积,部分肝中可见肉芽肿。100μg/kg剂量组血清SOD和GSH-Px活力升高,MDA含量降低,体现出CeO_(2)NPs的抗氧化作用,随着注射剂量的升高,CeO_(2)NPs的氧化应激作用增强。结论 低剂量CeO_(2)NPs对机体较安全,并表现出一定的抗氧化作用,随着剂量增加,CeO_(2)NPs对机体的毒性也逐渐增强。
Objective To investigate the effects of CeO_(2)NPs subacute exposure on body weight,organ structures,and redox indicators in male mice.Methods Thirty-five male ICR mice were randomly divided into five groups with seven mice in each group.Normal control and CeO2NP(100,500,1000 and 5000μg/kg)groups were included.Mice were injected with CeO_(2)NPs intraperitoneally every other day for 28 days.Results Compared with the control group,no significant difference was found in weight gain(P>0.05),but significant differences were observed in the liver coefficient,epididymal coefficient,and sperm survival rate(P<0.01).In the 5000μg/kg group,CeO_(2)NPs were deposited in the liver and spleen,and granuloma was found SOD and GSH-Px activities in serum of the 100μg/kg group were increased,while MDA content was decreased,which indicated the antioxidant effect of CeO_(2)NPs.With the increase in dose,oxidative stress induced by CeO_(2)NPs was enhanced.Conclusions Low-dose-CeO_(2)NPs are safe and exert an antioxidant effect.With the increase in dose,the toxicity of CeO_(2)NPs also increases gradually.
