摘要
以聚合草根为原料,采用超声波辅助乙醇法提取尿囊素。以尿囊素得率为考核指标,经单因素和正交优化试验得到的尿囊素提取工艺为乙醇浓度60%、超声功率300 W、提取温度60℃,尿囊素得率为0.85 mg/g。在0.1~0.3 mg/mL浓度范围内,尿囊素对DPPH自由基清除率随尿囊素浓度升高逐渐增加,当尿囊素浓度为0.3 mg/mL时,对DPPH清除率达到68%。1 mg/mL尿囊素对金黄色葡萄球菌(抑菌直径=39 mm)和大肠杆菌(抑菌直径=22 mm)的抑菌效果突出,而0.1 mg/mL尿囊素几乎没有抑菌作用。
Allantoin from roots of Symphytum of ficinale L was extracted by utrasonic assisted ethanol extraction process.Taking the yield of allantoin as the evaluation indicator,through single factor and orthogonal optimization experiments,the extraction process of allantoin was obtained as follows:ethanol concentration of 60%,ultrasound power of 300 W,extraction temperature of 60℃,and the yield of allantoin reached 0.85 mg/g.In the range of 0.1 to 0.3 mg/mL,the scavenging rate of DPPH free radicals by allantoin gradually increased with increasing concentration of allantoin,and when the concentration of allantoin was 0.3 mg/mL,the scavenging rate reached 68%.1 mg/mL allantoin had a prominent antibacterial effect on Staphylococcus aureus(antibacterial diameter=39 mm)and Escherichia coli(antibacterial diameter=22 mm),while 0.1 mg/mL allantoin had almost no antibacterial effect.
