摘要
建立可同时测定博落回中血根碱等四种生物碱类有效成分的HPLC方法,评价博落回不同部位生物碱有效成分含量的差异。采用Waters-Symmetry C18色谱柱(5μm,4.6 mm×250 mm),以乙腈-0.1%磷酸为流动相,梯度洗脱,流速为1.0 mL/min,波长284 nm,进样量5μL,柱温30℃。结果显示,血根碱、白屈菜红碱、原阿片碱、别隐品碱四种生物碱在一定浓度范围内线性关系良好,平均回收率范围分别在98.88%~99.73%、97.97%~99.20%、95.18%~99.86%、95.76%~100.48%。博落回的根、茎、叶中四种生物碱有效成分含量差异较大,根和叶中四种生物碱的含量远大于茎,说明根和叶是博落回合成生物碱类次级代谢产物的主要部位。叶中血根碱、白屈菜红碱的含量是根中含量的2倍。综上所述,建立的HPLC法具有简单、稳定、准确的特点,为博落回的合理有效利用提供参考。
The purpose of this research is to establish an HPLC method for the simultaneous determination of four alkaloid active ingredients in Macleaya cordata(Willd.)R.Br.,and to evaluate the content differences of alkaloids in different parts of Macleaya cordata(Willd.)R.Br..The chromatographic conditions were showed as follows:the column was Waters-Symmetry C18(5μm,4.6 mm×250 mm),the mobile phase was acetonitrile-0.1%phosphoric acid,the elution method was gradient elution,the flow rate was 1.0 mL/min,the UV detection wavelength was 284 nm,the sample volume was 5μL,and the column temperature was 30℃.The result showed that protopine,allocryptopin,sanguinarine and chelerythrine all possessed good linear relationship.The average recoveries of protopine,allocryptopin,sanguinarine and chelerythrine were 98.88%~99.73%、97.97%~99.20%、95.18%~99.86%、95.76%~100.48%,respectively.The content of the four alkaloids in the roots,stems and leaves was significantly different,and the content of the four alkaloids in the roots and leaves was much higher than that in the stems,indicating that the roots and leaves were the main parts of the secondary metabolites of alkaloids from Macleaya cordata(Willd.)R.Br..The content of sanguinarine and chelerythrine in leaves was 2 times that in roots.In summary,the established HPLC method was simple,stable and accurate,and provided a reference for the rational and effective use of Macleaya cordata(Willd.)R.Br..
