摘要
铁死亡是区别于细胞凋亡、细胞自噬、细胞坏死的一种新型细胞程序性死亡方式,由铁依赖性脂质过氧化驱动。其主要特征是细胞内游离铁的蓄积、活性氧(reactive oxygen species,ROS)的积累和铁依赖的膜结构脂质过氧化损伤。近年来研究表明,铁死亡在肝癌、结直肠癌、肾癌、卵巢癌、甲状腺癌等恶性肿瘤中有重要作用,而其在胃癌中的作用研究相对较少。本综述详细总结了铁死亡的发生和调控机制,阐述了胃癌治疗中铁死亡的诱导对于改善肿瘤耐药性所具的积极作用及诱导胃癌细胞铁死亡的潜在靶点。
Ferroptosis is a novel form of programmed cell death that is different from apoptosis,autophagy,and cell necrosis,driven by iron-dependent lipid peroxidation.Its main features are intracellular accumulation of free iron,accumulation of reactive oxygen species(ROS),and lipid peroxidative damage of iron-dependent membrane structures.In recent years,studies have shown that ferroptosis has an important impact on malignant tumors such as liver cancer,colorectal cancer,kidney cancer,ovarian cancer,and thyroid cancer.However,there are relatively few studies on gastric cancer.This article summarizes in detail the occurrence and regulation mechanism of ferroptosis,and expounds the research status and treatment prospect of ferroptosis in the treatment of gastric cancer.
