miRNA-485-5p靶向调控SOX5对结直肠癌细胞增殖、迁移、侵袭的影响分析

摘要 目的 分析miRNA-485-5p靶向调控性别决定区Y框蛋白5(SOX5)对结直肠癌细胞增殖、迁移、侵袭的影响。方法 取结直肠癌癌组织、癌旁组织,检测miRNA-485-5p、SOX5表达,购买结直肠癌结细胞株HT29,细胞培养后分为对照组、空载组、沉默组、过表达组,对照组常规培养,不做转染处理,空载组、沉默组、过表达组分别转染miRNA-485-5p-NC、miRNA-485-5pmimics、anti-miRNA-485-5p。鉴定转染效率,萤光素酶报告基因系统验证miRNA-485-5p、SOX5靶向关系,分析转染miRNA-485-5p对结直肠癌细胞生物学行为的影响。结果 结直肠癌癌组织中miRNA-485-5p表达量低于癌旁组织,SOX5表达量高于癌旁组织(P<0.05)。与对照组、空载组相比,沉默组miRNA-485-5p表达量降低,过表达组miRNA-485-5p表达量升高(P<0.05)。与对照组、空载组相比,沉默组SOX5表达量升高,过表达组SOX5表达量降低(P<0.05);与沉默组相比,过表达组SOX5表达量降低(P<0.05)。与对照组、空载组相比,沉默组细胞增殖活性、细胞迁移数、细胞侵袭数均升高,过表达组增殖活性、细胞迁移数、细胞侵袭数均降低(P<0.05);与沉默组相比,过表达组增殖活性、细胞迁移数、细胞侵袭数均降低(P<0.05)。与空载组相比,过表达A组、过表达B组增殖活性、细胞迁移、侵袭数降低,差异具有统计学意义(P<0.05);与过表达A组相比,过表达B组增殖活性、细胞迁移数、细胞侵袭数升高,差异具有统计学意义(P<0.05)。结论 miRNA-485-5p可抑制结直肠癌细胞增殖、迁移、侵袭,其作用机制可能为靶向调控SOX5表达,有望成为结直肠癌的治疗靶点。 Objective To analyze the effect of miRNA-485-5p targeting sex determining region Y box protein 5(SOX5)on proliferation,migration and invasion of colorectal cancer cells.Methods Expressions of miRNA-485-5p and SOX5 in colorectal cancer tissues and adjacent tissues were detected.The colorectal cancer cell line HT29 was purchased.Cells were divided into the control group,no-load group,silence group,and overexpression group.The control group was routinely cultured without transfection,while the no-load group,silent group,and overexpression group were transfected with miRNA-485-5p-NC,miRNA-485-5p mimics,and anti-miRNA-485-5p,respectively.The luciferase reporter gene system was used to identify the transfection efficiency and verify the targeting relationship between miRNA-485-5p and SOX5.The effect of miRNA-485-5p transfection on the biological behavior of colorectal cancer cells was analyzed.Results The expression of miRNA-485-5p in colorectal cancer was lower than that in adjacent tissues,while the expression of SOX5 was higher than that in adjacent tissues(P<0.05).Compared with the control group and the empty group,the expression of miRNA-485-5p in the silent group decreased,while the expression of miRNA-485-5p in the overexpression group increased(P<0.05).Compared with the control group and the empty load group,the expression of SOX5 in the silence group increased,while that in the overexpression group decreased(P<0.05);Compared with the silent group,the expression of SOX5 in the overexpression group decreased(P<0.05).Compared with the control group and the empty load group,the cell proliferation activity,cell migration number and cell invasion number in the silence group increased,while the proliferation activity,cell migration number and cell invasion number in the overexpression group decreased(P<0.05);Compared with the silent group,the proliferation activity,cell migration number and cell invasion number of the overexpression group decreased(P<0.05).Compared with the empty load group,the proliferation activity,cell migration and invasion number of overexpression group A and overexpression group B decreased significantly(P<0.05).Compared with overexpression group A,overexpression group B had a statistically significant increase in proliferation activity,cell migration and cell invasion(P<0.05).Conclusion miRNA-485-5p can inhibit the proliferation,migration and invasion of colorectal cancer cells,and its underlying mechanism could involve regulating the expression of SOX5,which may serve as a therapeutic target for colorectal cancer.
