样品储存温度及放置时间对血清补体C3、C4检测的影响

摘要 目的探讨样品储存温度及放置时间对血清补体C3、C4检测的影响,为规范临床样品的留取提供参考。方法利用散射比浊法对40例患者进行血清补体C3、C4水平进行检测。样品分装后置于18~25、2~8、-20℃条件下储存,分别在0、24、48、72、96、120、144、168h各测量一次血清补体C3、C4水平,比较数值变化情况。结果在18~25℃储存时,样品放置24、48、72、96、120、144、168h血清补体C3、C4检测结果与即刻检测相比差异均存在统计学意义(P<0.05),随着储存时间的延长,血清补体C3、C4检测结果逐渐升高;在2~8℃储存时,样品放置48h内血清补体C3检测结果与即刻检测相比差异无统计学意义(P>0.05),样品放置24h内血清补体C4检测结果与即刻检测相比差异无统计学意义(P>0.05);在-20℃储存条件下,样品放置48、96、120、144h血清补体C3、C4检测结果与即刻检测相比差异无统计学意义(P>0.05);在18~25、2~8℃及-20℃储存条件下,样品不同放置时间血清补体C3检测结果与即刻检测结果相比平均偏倚绝对值波动于7.46%~24.55%、0.52%~7.96%、0.47%~2.04%,血清补体C4检测结果与即刻检测结果相比平均偏倚绝对值波动于5.11%~23.34%、0.39%~6.16%、0.23%~2.20%。结论血清样品在不同储存温度及放置时间条件下,补体C3、C4检测结果存在明显变化,医学实验室可根据自身检测体系,合理评估不同放置时间及储存温度对血清补体C3、C4检测结果的影响,保证检验结果准确及有效性。 Objective To investigate the influence of storage temperature and duration on the detection results of serum C3 and C4 concentrations,in order to provide a reference for standardizing the collection and preservation of clinical specimens.Methods The concentrations of serum C3 and C4 among 40 patients were detected by nephelometry.These specimens were stored at 18-25℃,2-8℃and-20℃,and the concentrations of serum C3 and C4 were measured at 0h,24h,48h,72h,96h,120h,144h and 168h,respectively.Results Compared with immediate detection,the results of serum complement C3 and C4 were significantly different when the specimens were stored at 18-25℃for 24h,48h,72h,96h,120h,144h and 168h(P<0.05),and the results of serum complement C3 and C4 were gradually increased as the storage duration extended.Compared with immediate detection,when stored at 2-8℃,there was no statistically significant difference in serum C3 detection results within 48 hours(P>0.05),and there was no statistically significant difference in serum C4 detection results within 24 hours(P>0.05);Compared with immediate detection,there was no statistically significant difference in the detection results of serum complement C3 and C4 when the specimens were stored at-20℃for 48,96,120,and 144 hours(P>0.05);Compared to immediate detection results,the average absolute bias values of serum C3 detection results at different storage durations fluctuated between 7.46%to 24.55%,0.52%to 7.96%,and 0.47%to 2.04%,respectively.Under storage conditions of 18-25℃,2-8℃,and-20℃,the average absolute bias of serum C4 detection results fluctuated between 5.11%to 23.34%,0.39%to 6.16%,and 0.23%to 2.20%,respectively.Conclusion Serum C3 and C4 concentration detection results have significant changes under different storage temperatures and duration conditions.The laboratory can evaluate the influence of different storage durations and temperature on the detection results of serum C3 and C4 concentration according to its own detection system,so as to ensure the accuracy and validity of the test results.
