紫芽茶脱毒苗组织培养再生体系的建立

摘要 笔者以紫芽茶幼叶、叶柄和茎段为外植体,比较不同浓度激素NAA、6-BA、KT、IBA、2,4-D配比对不同组织诱导、分化等过程的影响。结果表明,紫芽茶外植体在75%乙醇和0.1%HgCl_(2)灭菌及1/2 MS培养条件下,1.0 mg·L^(-1)2,4-D与0.5 mg·L^(-1)KT组配处理的愈伤组织生长最佳,成活率为80%左右。以幼叶、叶柄和茎段作为紫芽茶外植体,通过适宜的激素处理,可以建立紫芽茶不同组织培养再生体系。 The young leaves,petioles and stem segments of Camellia sinensis var.kitamura were used as explants to compare the effects of different concentrations of hormone ratios of NAA,6-BA,KT,IBA and 2,4-D on the induction and differentiation of different tissues.The results showed that under the conditions of 75%alcohhol and 0.1%HgCl_(2) sterilization and 1/2 MS culture,1.0 mg·L^(-1)2,4-D and 0.5 mg·L^(-1)KT combined treatment of callus growth was the best,the survival rate was about 80%.With the young leaves,petioles and stem segments as the explants of Camellia sinensis var.kitamura,different tissue culture and regeneration systems of Camellia sinensis var.kitamura could be established through appropriate hormone treatment.
