海水中细菌的分离鉴定及其对动物源病原菌的抑菌活性研究

摘要 为了分离出具有抑菌活性的海洋细菌,利用M10培养基、P3培养基和BHI固体培养基从海水样品中分离细菌,采用固体琼脂打孔法从分离细菌中筛选对大肠杆菌、铜绿假单胞菌、金黄色葡萄球菌和单增李斯特氏菌有抑菌活性的细菌,PCR扩增具有抑菌活性的细菌的16S rDNA基因并进行测序分析。结果表明,从湛江地区海水中共分离出1615株细菌,其中246株具有抑菌活性,对4种指示菌均有抑菌效果的细菌有6株。对大肠杆菌抑菌效果最佳的菌株DB17-1抑菌圈直径为26 mm,对金黄色葡萄球菌抑菌效果最佳的菌株FP5-2抑菌圈直径为29 mm,对铜绿假单胞菌抑菌效果最佳的菌株AM20-8抑菌圈直径为20 mm,对单增李斯特氏菌抑菌效果最佳的菌株FM11-1抑菌圈直径为24 mm。具有抑菌活性的细菌共分为31个属,其中不动杆菌属和芽孢杆菌属最多,分别占21.5%和18.3%。结果提示,海水中存在具有良好抑菌效果的细菌,可为发掘新型抗菌药物提供参考。 In order to isolate marine bacteria with bacteriostatic activity,M10 medium,P3 medium and BHI solid medium were used to isolate bacteria from seawater samples,and bacteria with bacteriostatic activity against Escherichia coli,Pseudomonas aeruginosa,Staphylococcus aureus and Listeria monocytogenes were screened from isolated bacteria by solid agar punching,and 16S rDNA genes of bacteriostatic bacteria were amplified by PCR and sequence analysis was carried out.The results showed that a total of 1615 strains of bacteria were isolated from seawater in Zhanjiang area,of which 246 strains had antibacterial activity and 6 strains had antibacterial effect on all four index bacteria.The diameter of the inhibitory circle of the strain DB17-1 with the best antibacterial effect on Escherichia coli was 26 mm,the strain FP5-2 with the best antibacterial effect against Staphylococcus aureus was 29 mm in the inhibition circle diameter,the strain AM20-8 with the best antibacterial effect against Pseudomonas aeruginosa had a diameter of 20 mm,and the strain FM11-1 with the best antibacterial effect against Listeria monocytogenes had a diameter of 24 mm.The bacteriostatic bacteria were divided into 31 genera,among which Acinetobacter and Bacillus accounted for the most,accounting for 21.5%and 18.3%,respectively.There are bacteria with good antibacterial effect in seawater,and the research results can provide a theoretical basis for the discovery of new antibacterial drugs.
