摘要
为研究灵芝孢子油(GLSO)体内外抗氧化作用,试验采用体外测定GLSO对DPPH、ABTS、超氧自由基(·O^(-)_(2))的清除率,体内通过D-半乳糖构建小鼠衰老模型,测定衰老小鼠血清及肝组织中SOD、CAT、MDA含量和肝组织PI3K/AKT蛋白表达水平。结果显示,GLSO对DPPH、ABTS、超氧自由基(·O^(-)_(2))的清除作用相当于VC的84.05%、22.05%、46.48%;模型组的小鼠血清和肝组织与空白组相比,SOD、CAT值降低,MDA有所升高,p-AKT/AKT和p-PI3K/PI3K比值降低,同时各剂量间呈一定的依赖效应。研究结果表明,GLSO可能通过激活PI3K/AKT通路发挥抗氧化作用。
In order to study the antioxidant effects of Ganoderma lucidum spore oil(GLSO)in vivo and in vitro,the scavenging rates of DPPH,ABTS and·O^(-)_(2) free radicals by GLSO were measured in vitro,and D-galactose was used to construct a senescent mouse model in vivo.The contents of SOD,CAT and MDA in serum and liver tissue and the expression level of PI3K/AKT in liver tissue of senescent mice were determined.The results showed that the scavenging effect of GLSO on DPPH,ABTS and·O^(-)_(2) free radicals was equivalent to 84.05%,22.05%and 46.48%of that of VC.Compared with blank group,SOD and CAT in serum and liver of model group decreased,while MDA increased.The ratio of p-AKT/AKT and p-PI3K/PI3K decreased,and there was a certain dependence effect between different doses.The results suggested that GLSO might play an antioxidant role by activating the PI3K/AKT pathway.
