姜黄素对热应激条件下蛋鸡性腺机能和Sirtuins基因家族表达的影响

Effect of Curcumin on the Gonadal Function and Sirtuins Gene Family Expression in Laying Hens under Heat Stress

摘要 为探究姜黄素对热应激条件下蛋鸡性腺生理机能及Sirtuins基因家族表达量的影响,试验选用42只健康、体重相近的30周龄罗曼粉蛋鸡(公母各半),随机分为常温组(CK组)、高温组(HC组)和高温处理组(HT组),每组公鸡和母鸡各7只。常温组和高温组饲喂基础饲粮,高温处理组在基础饲粮中添加200mg/kg姜黄素。常温组保持温度在(25±3)℃,高温组和高温处理组在常温组的基础上采用循环高温,保持每天10:00~16:00温度(32±3)℃,各组湿度维持在65%~80%,热应激处理3周。结果显示,与常温组相比,高温条件对鸡的性腺产生不良影响,其产蛋率或采精量明显下降,差异显著(P<0.05);Sirtuins家族各基因在卵巢中表达量明显减少(P<0.05),而Sirt4、Sirt5和Sirt7在睾丸中表达量也显著降低(P<0.05)。添加姜黄素后,产蛋率或采精量均显著上升(P<0.05),卵巢中Sirt1、Sirt2、Sirt3、Sirt5基因和睾丸中Sirt4和Sirt5的表达量均显著上升(P<0.05)。研究结果表明,添加姜黄素能有效缓解热应激对鸡性腺机能和Sirtuins基因家族表达量的影响。 In order to explore the effect of curcumin on the gonadal physiology of laying hens and laying hens(21 males and 21 females)with similar body weight were randomly divided into room temperature group(CK group),high temperature group(HC group)and high temperature treatment group(HT group),with 7 males and females in each group.The normal temperature group and the high temperature group were fed the basic diet,and the high temperature treatment group was fed with basic diet 200 mg/kg curcumin.The normal temperature group maintained the temperature at 25±3℃,thehightemperaturegroupand thehightemperaturetreatmentgroup adopted cyclic high temperature,maintained the daily temperature of 10:00-16:00 at 32±3℃,the humidity of each group was maintained at 65%-80%,and the heat stress treatment was 3 weeks.The results showed that the gonads of chickens under high temperature conditions had adverse effects,and the laying rate or sperm harvest decreased significantly,and the difference was significant compared with the normal temperature group(P<0.05).Compared with normal temperature group,the expression levels of Sirtuins family genes in ovary were significantly decreased(P<0.05),and the expression levels of Sirt4,Sirt5 and Sirt7 in testis were also significantly reduced(P<0.05).After added curcumin,the egg production rate and semen volume increased significantly(P<0.05),and the genes expression levels of Sirtl,Sirt2,Sirt3,Sirt5 in ovary and Sirt4,Sirt5 in testis were significantly increased(P<0.05).The research results show that curcumin supplementation could effectively alleviate the effects of heat stress on gonadal function and Sirtuins gene family expression.
