基于GC-MS和HPLC测定沙棘籽油中脂肪酸、甾醇和黄酮类成分

摘要 目的鉴定沙棘籽油中脂肪酸、甾醇、黄酮类成分并进行含量测定。方法采用气相色谱-质谱联用(Gas Chromatography-Mass Spectrometry,GC-MS)法和高效液相色谱(High Performance Liquid Chromatography,HPLC)法测定沙棘籽油中脂肪酸、甾醇、黄酮类化合物。结果共鉴定出40个活性成分,其中脂肪酸23种、甾醇4种、黄酮13种。脂肪酸中油酸、亚油酸、亚麻酸含量较高,分别为226.581、398.478、244.694 mg/g,共占86.975%。4种甾醇成分分别为菜籽甾醇、豆甾醇、谷甾醇和Δ5-燕麦甾醇,其中谷甾醇和Δ5-燕麦甾醇含量较高,分别占总甾醇的88.086%和11.055%。检测出表没食子儿茶素、香橙素、芦丁、圣草酚、柚皮素、表儿茶素、花旗松素、槲皮素、异鼠李素、木犀草素、山柰酚、二氢杨梅素、杨梅素等13个黄酮类成分。结论沙棘籽油具有较高的营养价值,本研究为沙棘籽油的综合开发利用提供了研究基础。 Objective To identify and determine the fatty acids,sterols and flavonoids in seabuckthorn seed oil.Methods Gas chromatography mass spectrometry(GC-MS)and high performance liquid chromatography(HPLC)were used to determine fatty acids,sterols and flavonoids in seabuckthorn seed oil.Results A total of 40 active components were identified,including 23 fatty acids,4 sterols and 13 flavonoids.The contents of oleic acid,linoleic acid and linolenic acid in fatty acids were 226.581 mg/g,398.478 mg/g and 244.694 mg/g respectively,accounting for 86.975%.Four sterols including CAMPESTEROL,STIGMASTER-OL,sitosterol and DELTA5-AVENASTEROL were detected.The contents of sitosterol and DELTA5-AVENASTEROL were higher,accounting for 88.086%and 11.055%of total sterols respectively.Thirteen flavonoids including(-)-epigallocate-chin,4-chlorobenzoic acid,Rutin,Eriodictyol,Naringenin,L-Epicatechin,Taxifolin,Quercetin,Isorhamnetin,Luteolin,Kaempferol,Dihydromyricetin and Myricetin were detected.Conclusion The Seabuckthorn seed oil has high nutritive value.This study provides a basis for the comprehensive utilization of seabuckthorn seed oil.
