参萸养心汤对压力负荷型慢性心力衰竭大鼠的心脏保护作用及机制研究

Study on the cardioprotective effect and mechanism of Shenyu Yangxin decoction on rats with pressure load chronic heart failure

目的探究参萸养心汤对压力负荷型慢性心力衰竭(CHF)大鼠心脏功能的影响和机制。方法将30只SD雄性大鼠随机分为空白组、模型组和中药组(每组10只),其中空白组仅行腹主动脉穿线不缩窄,模型组和中药组均接受腹主动脉缩窄术。手术过程中空白组和中药组各死亡1只,模型组死亡2只。建模成功12周后,中药组按照13.2 g/(kg·d)的剂量喂养参萸养心汤(颗粒剂),空白组和模型组喂养同等剂量生理盐水。比较三组持续喂养30 d后的心电图状况、血流动力学及血清指标。结果中药组的P波振幅小于模型组,P波时限、QRS时限短于模型组,ST高度大于模型组(P<0.05)。中药组的左心室舒张末期压力(LVEDP)显著低于模型组,左室收缩压(LVSP)和左心室内压最大上升速率(+dp/dt_(max))显著高于模型组(P<0.05)。中药组的内皮素-1(ET-1)、丙二醛(MDA)水平显著低于模型组,超氧化物歧化酶(SOD)水平显著高于模型组(P<0.05)。结论参萸养心汤能有效改善压力负荷型CHF大鼠的心脏功能,这可能与改善血流动力学、微血管内皮功能和氧化应激作用等有关。

Objective To explore the effect and mechanism of Shenyu Yangxin decoction on the cardiac function of rats with pressure load chronic heart failure(CHF).Methods Thirty SD male rats were randomly divided into blank group,model group and Chinese medicine group(10 rats in each group),in which the blank group only received abdominal aortic threading without coarctation,and both the model group and the Chinese medicine group received abdominal aortic coarctation.During the operation,1 rat died in the blank group and the Chinese medicine group and 2 rats died in the model group.After 12 weeks of successful modeling,the Chinese medicine group was fed Shenyu Yangxin decoction(granules)at 13.2 g/(kg·d),and the blank group and the model group were fed the same dose of normal saline.The electrocardiogram,hemodynamics and serum indexes of the three groups were compared after 30 d of continuous feeding.Results The amplitude of P wave in the Chinese medicine group was less than that in the model group,the duration of P wave and QRS were shorter than those in the model group,and the ST height was greater than that in the model group(P<0.05).The left ventricular end-diastolic pressure(LVEDP)in the Chinese medicine group was significantly lower than that in the model group,the left ventricular systolic pressure(LVSP)and maximal rate of rise in blood pressure in the ventricular chamber(+dp/dt_(max))were significantly higher than those in the model group(P<0.05).The levels of endothelin-1(ET-1)and malondialdehyde(MDA)in the Chinese medicine group were significantly lower than those in the model group,and the level of superoxide dismutase(SOD)was significantly higher than that in the model group(P<0.05).Conclusion Shenyu Yangxin decoction can effectively improve the cardiac function of pressure load CHF rats,which may be related to improving hemodynamics,microvascular endothelial function and oxidative stress.
