摘要
WOX基因家族是植物特有转录因子,具有调控植物的生长发育、植物的胚胎发育、激素信号转导、抗逆代谢等作用。本研究利用桃全基因组数据,首次对桃全基因组中的WOX基因家族进行鉴定和生物学分析。结果表明:桃WOX基因家族共有67个成员,8条染色体上都有分布,不同成员的氨基酸数、分子量和等电点差异较大,大部分WOX蛋白富含酸性氨基酸且稳定性较差,所有WOX成员都为亲水性蛋白;桃WOX蛋白二级结构均由α-螺旋、β-折叠、延伸链和无规则卷曲四部分组成;桃WOX基因家族97%的成员都定位于细胞核中;系统进化树将桃WOX基因家族分为3个组,只有第Ⅰ组包含40个PpWOX蛋白和15个拟南芥WOX蛋白,其他两组都没有拟南芥的WOX蛋白;桃WOX基因结构中都含有外显子和内含子结构,但是数量差异较大;保守基序分析表明,共有10个保守基序(motif 1~motif 10)且所有WOX成员都含有motif 1,同一分支中motif的数目和种类更为相似而不同分支间有其特异的motif,与系统进化树分析具有一致性。该研究结果将为进一步分析WOX基因家族各个成员在桃的生长发育和抗逆胁迫等方面的应用提供一定的基础数据和参考依据。
TheWOX gene family is a plant specific transcription factor,which regulates plant growth and development,plant embryo development,hormone signal transduction,anti-adverse metabolism and other functions.In this study,WOX gene family in the whole peach genome was identified and biologically analyzed for the first time using the whole-genome data of peach.The results showed that:there are 67 members of the peach WOX gene family,which are distributed on 8 chromosomes.The amino acid number,molecular weight and isoelectric point of different members vary greatly.Most WOX proteins are rich in acidic amino acids and have poor stability,and all WOX members are hydrophilic proteins;the secondary structure of peach WOX protein consists of four parts:α-helix,β-sheet,extended strand and random coil;97%of the members of the peach WOX gene family are localized in the nucleus.The peach WOX gene family was divided into three groups by phylogenetic tree.Only groupⅠcontained 40 PpWOX proteins and 15 Arabidopsis WOX proteins,while the other two groups had no ArabidopsisWOX proteins;all the peachWOX gene structures contained exon and intron structures,but the number of them varies greatly.The conserved motif analysis showed that there were 10 conserved motifs(motif 1~motif 10)and all WOX members contained motif;the number and types of motifs in the same branch were more similar,while there were specific motifs among different branches,which was consistent with phylogenetic tree analysis.The results of this study will provide some basic data and reference for further analysis of the application of each member ofWOX gene family in the growth and development of peach and stress resistance.
