山西谷子地方品种微核心种质构建

摘要 构建山西谷子地方品种微核心种质,旨在为谷子目标性状精准鉴定与相关候选基因挖掘提供种质基础。在598份山西谷子核心种质的基础上,选取抽穗期、株高、节数、颈长、叶长、叶宽、穗长、穗粗、茎粗、单穗质量、穗粒质量、码数、码粒数、千粒质量和蛋白质含量等15个农艺、品质性状,使用QGAStation软件进行微核心种质构建抽样分析、全基因组重测序技术去除近缘种质,以构建微核心种质。结果表明,山西谷子地方品种微核心种质共包含322份种质资源。微核心种质与原始种质比较结果表明,各性状的均值与方差均没有表现出显著性差异,均值差异百分率与方差差异百分率均为0,极差符合率为98.36%,变异系数变化率为104.19%,遗传多样性指数变化率为94.81%。微核心种质较好地保留了原始种质的遗传变异,具有较高的变异度,同时具备较强的代表性。 Construction of minicore germplasm of Shanxi foxtail millet landraces aims to provide germplasm basis for identification of target traits and mining of candidate genes of foxtail millet.In this study,totally 15 agronomic and quality traits of 598 core germplasm of foxtail millet in Shanxi were uesd,including heading date,plant height,stem node number,neck length,leaf length,leaf width,panicle length,diameter of panicle,diameter of main stem,panicle weight,grain weight per panicle,branch number per panicle,grain number per branch,1000-grain weight,and protein content.QGAStation software was used to conduct sampling analysis of minicore germplasm construction,whole genome resequencing technology for removing closely related germplasm to construct minicore germplasm.The results showed that the minicore germplasm of foxtail millet landrace in Shanxi contained 322 accessions.The comparative results between minicore germplasm and original germplasm showed no significant difference in the mean value and the variance of the traits.The mean value difference(MD)and the variance difference(VD)were 0,the range coincidence rate(CR)was 98.36%,the coefficient of variation change rate(VR)was 104.19%,and the genetic diversity index change rate(GR)was 94.81%.The minicore germplasm retained the genetic variation of the original germplasm,and had a strong variation and representativeness.
