巴豆酰化效应蛋白在前列腺癌中的表达及其与肿瘤分期和分级的相关性

Analysis of crotonylated effector protein expression in prostate cancer and its correlation with tumour stage and grade

摘要 目的 探讨巴豆酰化效应蛋白ENL、AF9在前列腺癌中的表达水平以及与肿瘤分期分级和Gleason评分之间的相关性。方法 在GEPIA和Oncomine数据库中分析巴豆酰化效应蛋白ENL、AF9在前列腺癌中的表达情况;对包含不同分期分级的前列腺癌标本130例和正常前列腺组织31例的组织芯片进行免疫组织化学染色,检测ENL、AF9在各样本中的表达水平,并探究ENL、AF9的蛋白表达水平与前列腺癌分期分级和Gleason评分的关系。结果 与正常组织相比,ENL的mRNA表达水平和蛋白表达水平在前列腺癌中明显升高,且差异具有统计学意义。与正常组织相比,前列腺癌中AF9的mRNA表达水平和蛋白表达水平变化不明显,差异无统计学意义;ENL的蛋白表达水平与前列腺癌的分期和分级无明显相关性,与前列腺癌的Gleason评分呈正相关,且ENL的表达水平更高的前列腺癌患者的无疾病进展期生存期(Disease free survival, DFS)更短,差异具有统计学意义。结论 ENL在前列腺癌组织中显著高表达,与前列腺癌的恶性进展(Gleason评分)相关,可作为前列腺癌的诊断标志物之一,将来有可能作为前列腺癌的治疗靶点。 Objective To investigate the expression levels of the crotonylation effector proteins ENL and AF9 in prostate cancer and their correlation with tumour stage and Gleason score.Methods Using the GEPIA and Oncomine databases,we analysed the expressions of the crotonylation effector proteins ENL and AF9 in prostate cancer;subsequently,we employed immunohistochemistry to detect the expressions of ENL and AF9 in a tissue microarray derived from 130 cases of prostate cancer of different stages and 31 cases of normal prostate tissue.and explored the relationship of the protein expression levels of ENL and AF9 with prostate cancer stage grading and Gleason score.Results Compared with normal tissues,the mRNA expression and protein expression levels of ENL were all significantly increased in prostate cancer tissues.The mRNA expression and protein expression levels of AF9 were not obviously altered in prostate cancer tissues compared with normal tissues;the protein expression level of ENL was not correlated with the stage and grade of prostate cancer,but was positively correlated with the Gleason score of prostate cancer.The patients with high expression of ENL were found to have shorter disease⁃free survival(DFS)compared with the patients with low expression of ENL.Conclusion ENL was highly expressed in prostate cancer tissue and correlated with prostate cancer progression(Gleason score).It may become a diagnostic and potential therapeutic target for prostate cancer in the future.
