黑藻多肽亚铁螯合物的制备及其抗氧化活性研究

摘要 该文以黑藻多肽为原料制备黑藻多肽亚铁螯合物,探索温度、pH、时间、多肽与氯化亚铁质量比、抗坏血酸添加量对黑藻多肽与亚铁螯合率的影响.采用紫外-可见分光光度法(UV-vis)和傅里叶红外光谱法(FT-IR)对螯合物的结构进行表征,并考察了螯合物抗氧化活性.结果表明:多肽和亚铁最佳螯合条件为温度30℃、pH 7.0、时间30 min、多肽和氯化亚铁质量比为8∶1、抗坏血酸添加量4.5 mg/mL,在此条件下螯合率为64.53%;UV-vis和FT-IR的光谱图分析证实黑藻多肽亚铁螯合物已被成功制备,且多肽的羟基、氨基、羧基和酰胺键参与了黑藻多肽与亚铁的螯合反应;螯合物对1-二苯基-2-三硝基苯肼自由基(DPPH·)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS^(+)·)和羟基自由基(·OH)的IC_(50)值分别为66.06μg/mL、27.77μg/mL和125.05μg/mL,相比黑藻多肽,螯合物对这3种自由基的清除能力分别提高了42 268.42%,78.63%和5.61%.本研究结果为黑藻多肽的综合利用提供理论基础. In this work,the chelate of polypeptide from Hydrilla verticillata(Linn.f.) Royle(HVR) and ferrous was prepared.And the effects of temperature,pH,time,mass ratio of polypeptide to FeCl_(2) and the amount of V_(C) on chelating rate were investigated.The structure of the chelate was characterized by UV-vis and FT-IR,and its antioxidant activity in vitro was investigated.The results showed that the optimal chelation conditions were as followed:temperature 30 ℃,pH 7.0,30 min,mass ratio of polypeptide to FeCl_(2) 8:1 and ascorbic acid 4.5 mg/mL,under this condition,the maximum chelating rate was 64.53%.Uv-vis and FT-IR analysis were employed to confirmed successful preparation of chelate,and hydroxyl,amino,carboxyl amide bonds of the polypeptide were involved in chelation reaction.The IC_(50 )valuesof chelate to DPPH·,ABTS^(+)·and ·OH were 66.06 μg/mL,27.77 μg/mL and 125.05 μg/mL,respectively,the scavenging rates of chelate to the above-mentioned three radicals were higher than polypeptide with 42 268.42%,78.63% and 5.61%,respectively.The results of this study provided theoretical basis for the utilization of HVR polypeptide.
