RGS16在胃癌中的表达及临床意义

Expression and clinical significance of RGS16 in gastric cancer

目的 检测胃腺癌中G蛋白信号调节16(RGS16)的表达,探讨其与胃腺癌患者临床特征的关系。方法 选取2016年1月-2016年12月盐城市第一人民医院普外科的88例胃癌患者为研究对象。运用免疫组织化学法和Western Blot检测患者癌及癌旁组织中RGS16蛋白的表达水平。分析RGS16表达水平与组织病理参数之间的关系,使用Kaplan-Meier和Log-rank检验进行生存分析,并采用COX风险比例模型分析影响胃癌预后的因素。结果 癌组织中RGS16的过表达率(61.40%)高于癌旁组织(28.40%),差异具有统计学意义(P<0.01)。Western blot结果显示,与癌旁组织相比,RGS16在癌组织中表达更高(P<0.001)。RT-qPCR检测结果显示,RGS16在癌组织中的mRNA表达水平(5.97±4.88)较癌旁组织(2.19±1.99)高,差异具有统计学意义(P<0.01)。Kaplan-Meier生存函数结果:RGS16低表达的患者5年生存率显著高于RGS16高表达的患者(P<0.001)。RGS16蛋白的高表达和肿瘤的浸润深度是胃癌不良预后的危险因素(P<0.001)。结论 RGS16的高表达是影响胃癌患者术后不良预后的危险因素,可能为胃癌早期诊断和靶向治疗提供依据。

Objective To detect the expression of G protein signaling regulation 16(RGS16)in gastric cancer(GC)and explore its relationship with the clinical characteristics of GC patients.Methods Eighty-eight patients with GC in the General Sur-gery Department of the First People's Hospital of Yancheng from January 2016 to December 2016 were selected as the study subjects.RGS16 protein levels in cancerous and paracancerous tissues were detected by immunohistochemistry and Western blot.Chi-square test was used to analyze the relationship between RGS16 and histopathological parameters.Kaplan-Meier and Log-rank tests were used for survival analysis.Cox proportional-hazards model was used to analyze the factors affecting the prognosis of GC.Results Compared with paracanceroustissues,the expression of RGS16 in cancerous tissues was significantly increased(P<0.01).Further analysis indica-ted that the high expression of RGS16 was significantly correlated with lymphatic metastasis,depth of infiltration and tumor differentia-tion(P<0.05).Conclusion The high expression of RGS16 is an independent risk factor affecting the poor prognosis of GC patients af-ter surgery,and may provide a basis for early diagnosis and targeted therapy of GC.
