摘要
目的:优化超声辅助双水相法制备茯苓提取物的工艺。方法:选取茯苓中去氢土莫酸、茯苓酸A、松苓新酸为目标成分,经层次分析法量化三者产率权重,对其进行综合评分。以综合评分为响应值,通过响应面法优化超声辅助双水相体系制备茯苓提取物的工艺。结果:乙醇-NaH_(2)PO_(4)双水相体系成相能力好,提取效率高,可用于制备茯苓提取物。最佳工艺为乙醇质量分数34%,NaH_(2)PO_(4)质量分数18%,液固比19 mL/g,此时综合评分为90.82。结论:该方法条件温和、提取率高且操作简单,可用于茯苓提取物的制备。
Objective:Optimize the preparation technology of Poria cocos extraction using ultrasound assisted aqueous two-phase system.Methods:Dehydrotumulosic acid,poricoic acid A and dehydrotrametenolic acid in Poria cocos are selected as the target components.The analytic hierarchy process is developed to weight the yield of these three components and the comprehensive weighted score is used as the response value.The preparation method of Poria cocos extraction based on ultrasound assisted aqueous two-phase system is optimized through the response surface methodology.Results:Ethanol and sodium dihydrogen phosphate(NaH_(2)PO_(4))are chosen to construct the aqueous twophase system due to the strong phase-separation ability and high extraction rate.The optimum condition is determined as follows:ethanol mass fraction of 34%,NaH_(2)PO_(4) mass fraction of 18%,liquid-solid ratio of 19 mL/g.Under the above condition,the comprehensive weighted score reaches 90.82.Conclusion:The proposed method has the advantages of mild condition,high extraction rate,easy operation and shows a promising prospect in preparing Poria cocos extraction.
