黄连素干预牙周炎小鼠对炎症基因和牙槽骨吸收的影响

摘要 目的:探讨黄连素干预牙周炎小鼠对病灶局部炎症基因表达和牙槽骨吸收的影响。方法:21只C57BL/6雄性小鼠,随机分为对照组(健康小鼠)、模型组(牙周炎小鼠)和干预组(黄连素连续干预4周),每组7只小鼠,观察各组小鼠一般情况、组织切片的变化、牙槽骨的吸收程度,同时用荧光定量PCR检测牙周组织炎症因子IL-1β、TNF-α、IL-10等基因的表达情况。结果:经黄连素治疗后,干预组牙周情况得到显著改善,干预组小鼠的牙槽骨吸收量低于模型组;干预组的炎症因子基因IL-1β、TNF-α、IL-10表达水平较模型组降低,组间比较差异均有统计学意义(P<0.05)。结论:黄连素对牙周炎的发生发展有显著抑制效果,且能在一定程度上改善牙槽骨的吸收情况。 Objective:To investigate the inhibitory effect of berberine on periodontitis and dental plaque growth.Methods:Twenty-one C57BL/6 male mice are randomly divided into control group(healthy mice),model group(periodontitis mice)and intervention group(berberine is intervened continuously for 4 weeks),7 mice in each group.The general condition,the changes of tissue sections and the degree of alveolar bone absorption are observed.At the same time,the expression of inflammatory factors IL-1β,TNF-αand IL-10 genes in periodontal tissues is detected by fluorescence quantitative PCR.Results:After berberine treatment,the periodontal condition of the intervention group is significantly improved,and the alveolar bone absorption of the intervention group is lower than that of the model group.The levels of inflammatory factor genes IL-1β,TNF-αand IL-10 in the intervention group are lower than those in the model group,and the differences between groups are statistically significant(P<0.05).Conclusion:Berberine can significantly inhibit the occurrence and development of periodontitis and dental plaque,and can improve the resorption of alveolar bone to a certain extent.
