老年人接种甲型流感疫苗前后在不同衰弱状态下基因表达变化 被引量:1

摘要 目的通过对转录组测序数据分析,探索老年人接种甲型流感疫苗前后在不同衰弱状态下基因表达变化。方法从GEO数据库中下载转录组测序数据,分别计算衰弱与非衰弱老年人在接种疫苗前和接种后3、7 d的差异表达基因,并使用GO功能富集分析和KEGG通路分析对差异基因进行功能注释;进一步根据差异基因的相关性,当设定关联系数阈值为0.85时,参数β=5,基于此创建基因共表达网络;分选出与临床表型呈显著性相关的基因模块,最终根据模块内部的相连性筛选出核心枢纽基因。结果衰弱和非衰弱老年人在接种疫苗前和接种后3、7 d,分别有2756、1558、1783个差异表达基因。依据差异基因表达的相关程度,共分选出12个基因共表达模块,其中绿色模块与年龄及衰弱状态表型呈显著相关;发现B3GAT1、PTGDR等17个基因位于共表达网络的核心区;生物注释功能结果进一步表明,模块内基因主要富集于适应性免疫应答、T细胞激活、NK细胞激活等生物学过程中。结论不同衰弱状态的老年人在接种甲型流感疫苗前后基因表达,免疫反应、T细胞激活等信号通路存在差异。 Objective Through transcriptome sequencing data analysis,explore the changes in gene expression in elderly individuals with different frailty statuses before and after receiving influenza A vaccine.Methods We downloaded transcriptome sequencing data from the GEO database and analyzed the differentially expressed genes(DEGs)between frail and non-frail groups at Days 0,3 and 7 post-vaccination.GO functional enrichment analysis and KEGG pathway analysis were used to annotate the functions of the DEGs.According to the correlation of genes,the parameterβ=5 was used to construct the gene co-expression network when the correlation coefficient threshold was set to 0.85 and screened gene modules significantly associated with clinical traits.The hub genes were screened according to the connectivity within modules.Results There were 2756,1558,and 1783 differentially expressed genes between frail and non-frail older adults before vaccination and 3 and 7 days after vaccination,respectively.A total of 12 gene co-expression modules were constructed from the correlations of gene expression,in which the green modules were significantly correlated with the Age and Frailty status;We found that 17 genes including B3GAT1,PTGDR were in the hub of the co-expression network;The result of biological annotation function revealed that the genes in green modules were mainly enriched in adaptive immune response,T cell activation and natural killer cell activation pathways.Conclusion Gene expression in elderly individuals with different frailty statuses changes before and after receiving influenza A vaccine,with differences observed in immune response,T cell activation,and other signaling pathways.
